趋化因子CCL20、SDF-1在大肠癌中的表达及其与淋巴结及肝脏转移的关系

【摘要】 目的探讨趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4在大肠癌中的表达及与淋巴结及肝脏转移的关系。方法选取结直肠癌患者96例,采用荧光定量反转录聚合酶链反应(QRTPCR)方法检测96例行大肠癌根治术患者的大肠癌组织、正常肠黏膜组织及其中18例大肠癌肝转移组织及肝转移灶旁肝组织中趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4 mRNA表达情况。结果趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4在结直肠癌肝转移和肝转移中的高表达,与正常肠黏膜组织相比较,差异有统计学意义(P <0. 05)。趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4在大肠癌肝转移旁组织与正常肠黏膜组织相比较,差异无统计学意义(P> 0. 05)。趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4与患者性别、年龄、肿瘤分期差异无统计学意义(P> 0. 05)。趋化因子CCL20及其受体CCR6、SDF-1及其受体CXCR4与淋巴结转移及远处转移有关,差异有统计学意义(P <0. 05)。结论 CCL20及其受体CCR6、SDF-1及其受体CXCR4在大肠癌中高表达,可能与大肠癌的进展和转移有关。

【Abstract】 Objective To investigate the expression of chemokine CCL20 and its receptor CCR6,SDF-1 and its receptor CXCR4 in colorectal cancer and their correlations with lymph node and liver metastasis. Methods Ninety-six patients with colorectal cancer admitted from June 2015 to June 2018 were selected. The colorectal cancer,adjacent tissues of colorectal carcinoma and normal intestinal mucosa tissues were collected,while liver metastatic tissues were collected in 18 cases. Fluorescence quantitative reverse transcriptase polymerase chain reaction( QRT-PCR) was applied for assessment of the expression of CCL20,CCR6,SDF-1 and CXCR4 mRNA. Results The expression of CCL20,CCR6,SDF-1 and CXCR4 in liver metastatic tissues and colorectal cancer tissues in those with liver metastasis was significantly higher than that in normal intestinal mucosa( P < 0. 05). There was no significant difference in CCL20,CCR6,SDF-1 or CXCR4 between adjacent tissues of colorectal carcinoma and normal intestinal mucosa( P > 0. 05). There was no significant difference in chemokine CCL20,CCR6,SDF-1 or CXCR4 among genders,ages or tumor stages( P >0. 05). The expression of CCL20,CCR6,SDF-1 and CXCR4 was significantly associated with lymph node metastasis and distant metastasis( P < 0. 05). Conclusion The high expression of CCL20,CCR6,SDF-1 and i CXCR4 is correlated to the progression and metastasis of colorectal carcinoma.
