基于EST-SSR标记的部分玉兰属植物亲缘关系分析

【摘要】 为研究部分玉兰属植物的亲缘关系,利用EST-SSR标记对21个玉兰种(品种)进行遗传多样性以及亲缘关系分析。本研究共筛选得到30对多态性引物,多态扩增率为73.17%,平均Nei基因多样性(H)和Shannon’s信息指数(I)分别为0.56和0.93,表明玉兰属种质资源的遗传多样性较丰富。通过NTSYS软件进行相似性系数计算,21个玉兰种(品种)的相似系数分布在0.55~0.84之间。UPGMA聚类结果和主坐标分析结果基本一致,即可将21份材料分为4大类:二乔玉兰类群、白玉兰类群、望春玉兰类群及渐叶玉兰类群。本研究为玉兰属种质资源的遗传图谱绘制和新品种选育等提供技术服务及理论指导。

【Abstract】 To study the genetic relationship among some Yulania Spach. plants, EST-SSR markers were used to analyze the genetic diversity and relationships of 21 Yulania Spach. species(cultivars). 30 pairs of polymorphic primers were screened out in this study, with the polymorphism rate of 73.17%. The average of Nei’s genetic diversity(H) and Shannon’s information index(I) were 0.56 and 0.93, respectively, indicating that the germplasm resources of Yulania Spach. had a high genetic diversity. The similarity coefficients of 21 Yulania Spach. species(cultivars) were between 0.55 and 0.84, which were calculated by NTSYS software. The result of UPGMA cluster analysis was basically similar with the principal coordinate analysis, that was the 21 materials could be divided into 4 groups: Magnolia soulangeana Soul.-Bod. taxa Y. denudate taxa, Y. biondii taxa, and Magnolia acuminate taxa. This study could provide technical help and theoretical guidance for the mapping of genetic maps and new variety breeding of Yulania Spach. germplasm resources.
