利用SLAF-seq技术构建水稻籼粳交DH群体遗传图谱
Construction of Genetic Map for Rice Doubled Haploid Population by Specific-length Amplified Fragment Sequencing
【摘要】 我们利用特定位点扩增片段测序技术(specific-length amplified fragment sequencing,SLAF-seq)对籼稻窄叶青8(ZYQ8)和粳稻京系17(JX17)构建的双单倍体(doubled haploid,DH)群体进行了简化基因组测序。将DH群体测序数据与水稻参考基因组进行比较分析,获得了70 997个在两个亲本中存在多态性的SLAF标签。通过进一步的筛选,我们利用8 940个高质量的SLAF标签构建了该群体的遗传图谱。图谱覆盖12个连锁群,总图距为2 294 71 c M,标记间的平均图距为0.26 c M。遗传图谱的连锁群与水稻物理图的相关系数统计显示,除了9号连锁群的相关系数为0.87外,其余的11个连锁群的相关系数均在0.95以上,表明该图谱的12个连锁群与水稻物理图有很好共线性关系。该高密度图谱的构建为我们进一步研究籼、粳差异基因提供了帮助。
【Abstract】 A doubled haploid(DH) population constructed by an indica cultivar ZYQ8 and a japonica cultivar JX17 was sequenced by specific-length amplified fragment Sequencing technology. A total of 70 997 SLAF markers were obtained by comparing the sequenced data with rice reference genome. The genetic map for the DH population was constructed by 8 940 selected high quality SLAF markers. The genetic map covered 12 linkage groups and was 2 294 71 c M in length with an average distance of 0.26 c M between adjacent markers. The statistic analysis between the linkage groups of the genetic map and the rice physical map showed that the correlation coefficient of the chromosome 9 was 0.87, and correlation coefficients of the other eleven chromosomes were all above 0.95, indicating a great collinearity relationship between the 12 linkage groups of the genetic map and the rice physical map. The high density genetic map might provide a platform for further study of specific genes in indica and japonica cultivars.
