藏族、白族18个面部相关SNPs位点多态性及群体差异性研究
Genetic polymorphism and genetic structure difference of 18 facial related SNPs loci in Tibetan and Bai ethnic groups
【摘要】 目的获取18个面部相关SNPs位点在藏族人群和白族人群的遗传多态性,探索不同民族群体间遗传结构的差异性。方法采用SNa Pshot技术建立18-SNPs位点复合检测体系并检测192名藏族人群和159名白族人群18个SNPs位点的基因型。应用SPSS23.0软件进行统计学分析,R v3.3软件进行主成分分析。结果除了rs987525位点仅检测出一种等位基因G,其余位点的基因分型均具有很好的多态性,其中rs502393、rs7559271、rs920683、rs974448、rs6555969、rs13267109、rs2724626、rs1258763、rs1978860、rs2788888、rs934498 11个SNPs位点的等位基因频率在两个民族群体间差异有显著统计学意义(P<0.01),rs502393、rs7559271、rs920683、rs974448、rs642961、rs13267109、rs2724626、rs1978860、rs2788888、rs934498 10个SNPs位点的基因型频率在两个民族群体间差异有显著统计学意义(P<0.01),主成分分析(PCA)散点图中4个群体相互独立分布(云南傣族、北京汉族群体数据来自Ensembl千人数据库),群体间遗传结构差异明显。结论藏族、白族18个面部相关SNPs位点的基因分型具有很好的多态性,丰富了我国少数民族面部相关SNPs位点的数据库,其人群结构差异性在法医学应用具有重要的意义。
【Abstract】 Objective To obtain the genetic polymorphism of 18 facial related SNPs loci in Tibetan and Bai ethnic groups,and to explore the difference of genetic structure among different ethnic groups.Methods An 18-plex SNP assay was established and SNa Pshot assay was used to analyze the genotype of 18 SNPs loci in 192 Tibetan ethnic group and 159 Bai ethnic group. The SPSS23. 0 software was used for statistical analysis,and the principal component analysis was performed by R v3. 3 software. Results Except only rs987525 locus was detected for only one kind of allelic gene,the others revealed a great genetic polymorphism. The allele frequency of rs502393,rs7559271,rs920683,rs974448,rs6555969,rs13267109,rs2724626,rs1258763,rs1978860,rs2788888 and rs934498 were significantly different between the two groups( P < 0. 01). The genotypic frequency of rs502393,rs7559271,rs920683,rs974448,rs642961,rs13267109,rs2724626,rs1978860,rs2788888 and rs934498 were significantly different between the two groups( P < 0. 01). Among the four groups( Yunnan Dai,Beijing Han data from the Ensembl thousand people’s database),Principal-component analysis showed that the genetic structure were significantly different between the groups. Conclusion The 18 SNPs loci revealed a great genetic polymorphism in Tibetan and Bai ethnic group, This study has enriched the Chinese ethnic minorities facial related SNPs loci database,and its population genetic structure differences have great significance in forensic medicine.
