恒河猴牙髓干细胞的牙向分化
Tooth Differentiation of Rhesus macaques Dental Pulp Stem Cells
【摘要】 将恒河猴牙髓剪碎,用酶消化后培养获得恒河猴牙髓干细胞,用细胞免疫荧光的方法鉴定干细胞表面标记基因的表达,对分离的细胞进行成脂成骨的诱导,同时将细胞与支架混合后移植裸鼠皮下.结果发现,分离的细胞能持续传代,能被条件性地诱导成脂和成骨,能表达CD146,OCT4,SSEA4,STRO1等干细胞的标记基因,细胞与支架移植体内能分泌形成牙本质.结果表明酶消化法能够成功分离恒河猴的牙髓干细胞,该细胞在体内能分化形成牙本质.实验结果为该细胞进一步作为种子细胞用于牙齿再生研究奠定了实验基础.
【Abstract】 Cutting up the dental pulp of Rhesus macaques and harvesting the Rhesus macaques DPSCs( r DPSC) after enzymes digestion. Identifying the r DPSCs by investigating the expression of the surface marker genes of stem cells with immunofluorescence. Performing adipogenic and osteogenic induction. Mixing the r DPSCs and HA/TCP and transplant them subcutaneously in nude mice. The results indicated that the r DPSCs were separated successfully and can be passaged continuously. The r DPSCs expressed CD146,OCT4,SSEA4,STRO1 which are the marker gene of stem cells. r DPSCs produce to dentin after transplanting to vivo. This research separated and identified the r DPSCs,setting a foundation for the tooth regeneration via these stem cells as seed cell.
