香椿组培增殖和生根培养基筛选

【摘要】 为研究香椿组培快繁技术,对香椿组织培养的培养基配方进行筛选。在MS基本培养基中添加不同浓度的6-BA、NAA、GA3的正交试验结果表明,影响香椿芽增殖的主要因素是6-BA,且对芽增殖的影响达显著水平(P<0.05),最佳质量浓度为1.0 mg·L-1;对芽增殖影响次要因素是NAA,最佳质量浓度为0.1 mg·L-1;GA3对芽增殖的影响最小,但对幼茎节间伸长生长有促进作用;芽增殖最佳培养基为MS+6-BA 1.0 mg·L-1+NAA 0.1 mg·L-1+GA30.1 mg·L-1,芽增殖倍数可达3.6倍。以MS、1/2 MS为基本培养基添加不同浓度的NAA、IBA进行的生根培养试验结果表明:MS+NAA 0.2mg·L-1+IBA 0.2 mg·L-1培养基生根效果最好,生根率可达25%。腐殖土作为香椿组培苗移栽基质,组培苗移栽成活率可达95%,苗木长势好。

【Abstract】 In order to study the tissue culture and rapid propagation of Toona sinensis,the medium formula for tissue culture of Toona sinensis was studied and screened.By adding different concentrations of 6-BA,NAA and GA3 in MS medium,the orthogonal experiment and variance analysis showed that the main factor affecting the proliferation of Toona sinensis was 6-BA,which had a significant effect on the bud proliferation(P < 0.05),and the best concentration was 1.0 mg·L-1;the secondary factor affecting bud proliferation was NAA,and the optimal concentration was 0.1 mg·L-1;GA3 had the least effect on bud proliferation,but GA3 had a promoting effect on the internode elongation growth of young stems.The optimal bud culture medium combination was MS + 6-BA 1.0 mg·L-1+ NAA 0.1 mg·L-1+ GA30.1 mg·L-1,and the multiplication ratio of shoots reached 3.6 times.MS and 1/2 MS medium supplemented with different concentrations of NAA and IBA to carry out rooting culture test,the results showed that the MS + NAA0.2 mg·L-1+ IBA 0.2 mg·L-1 formula combination medium had the best rooting effect,and the rooting rate was 25%.Taking humus soil as tissue culture seedlings transplanting toothache,the survival rate of the tissue culture seedlings transplanted was high up to 95%,and the seedlings grew well.
