三维追踪方法用于HEP-2细胞的受体介导成像
Three-dimensional tracking method for receptor mediated imaging of HEP-2 cells
【摘要】 本文基于双螺旋点扩散函数方法,建立了具有高定位精度的三维多粒子追踪成像系统,成像深度达到4μm,利用荧光珠进行标准实验,通过探测探针在甘油-水溶液中的均方位移曲线得到了粒子扩散系数。最后利用本系统实现了对喉癌(HEP-2)细胞内多个荧光微球的动态追踪.通过数据分析得到细胞内荧光微球的移动量以及运动速率,为研究活细胞内分子转运和相互关系提供了非常有用的工具.
【Abstract】 We established a highly accuracy three-dimensional particle tracking imaging system based on double-helix point spread function,with 4 μm imaging depth. According to our system,the diffusion coefficients were obtained by detecting the mean square displacement curves of particles in glycerol water solution. As a useful tool for studying the molecular transport and interaction in living cells,the dynamic tracking of multiple fluorescent microspheres in laryngeal carcinoma(HEP-2) cells was realized by our system. The amount and velocity of intracellular fluorescent microspheres were obtained through data analysis.
