壳聚糖包覆水滑石药物载体的制备及缓释性能

【摘要】 选用壳聚糖和镁铝层状双羟基氢氧化物(水滑石)为材料,采用自组装包埋的方法制备核壳结构的复合药物载体微球。分别用以乳化交联法制备的壳聚糖微球与以共沉淀法制备的水滑石在一定条件下生成壳聚糖微球包埋水滑石的复合结构载体。所用模拟药物为具有抗炎作用的黄芩素。利用X射线衍射分析(XRD)、红外(FT-IR)、透射电镜(TEM)、扫描电镜(SEM)、药物体外释放等方法对最终合成的材料进行表征与缓释性能分析,结果表明,黄芩素在十二烷基硫酸钠(SDS)胶束的包裹下插入到水滑石层间,形成载药水滑石,再整体被壳聚糖微球所包埋形成核壳结构的复合载药微球。通过这种复合药物载体层层包覆药物的形式有效地提高了模拟药物黄芩素的缓释性能。

【Abstract】 In this work,chitosan( CS) and Mg Al-layered double hydroxides( LDHs) was used as predecessors to develop a novel composite core-shell drug carrier by self-assembly and embedding. Generally,CS microspheres were synthesized by reversed-phase microemulsion method,and LDHs were synthesized by a simple co-precipitation method. The microparticles were composed of chitosan microspheres and layered double hydroxides-drug. Baicalein was used as the simulation drug in this paper. The structures were characterized by X-ray diffraction( XRD),Fourier transform infrared spectrometer( FTIR spectrometer),transmission electron microscope( TEM) and scanning electron micrograph( SEM). The results showed that the SDS micelle with baicalein was intercalated into interlayer. Mg Al-layered double hydroxides with baicalein coated with chitosan microsphere. Hybridization of chitosan microspheres and baicalein intercalated Mg Al-layered double hydroxides could provide a suitable drug delivery system.
