【摘要】 为建立快速、可靠的评估牛囊胚质量的双重染色方法,对目前已建立的4种双重染色方案进行了改进。结果显示:改进的方案一、方案三及方案四都能达到牛囊胚双重染色的效果;改进的方案三、方案四囊胚细胞总数与Hoechst33342染色结果无差异(84.33个±9.98个VS 83.63个±9.74个,P>0.05;93.00个±10.50个VS 83.63个±9.74个,P>0.05),而且囊胚内细胞团(ICM)数与囊胚细胞总数的比值分别为0.338 2与0.335 6,均接近标准值0.33;改进的方案四染色最为稳定,效果较好,整个染色时间由原来的12 h(过夜)降到不到3 min,极大地提高了染色效率,可以作为一种快速、简洁、有效的牛囊胚质量鉴别方法。更多还原

【Abstract】 This study aimed to establish a quick and reliable double-staining method for assessing the quality of blastocyst produced by in vitro fertilization. The four common used methods were modified. The results indicated that the improved method1,3,4 realized double-staining of blastocysts. Almost the same total blastocyst cell numbers were gotten by the improved method3,4 and Hochest 33342 staining respectively(84.33±9.98 VS 83.63±9.74,P >0.05;93.00±10.50 VS 83.63±9.74,P >0.05).And the ratioes of number of cells in inner cell mass to total blastocyst cells were 0.338 2 and 0.335 6 respectively,which were very close to the standard value(0.33). The improved method 4 not only showed stable and effective result,but also a very short staining time,decreased greatly from more than 12 h(overnight) to 3 min,compared to original method. So the modified method 4 improved staining efficiency greatly and may be used as a quick,simple and effective method for assessing blastocysts quality.更多还原
