商陆总皂苷对四氯化碳诱导的小鼠急性肝损伤的作用
Protective effect of the total saponins from Phytolacca acinosa against carbon tetrachloride induced acute liver injury in mice
【摘要】 目的:研究商陆总皂苷对四氯化碳(CCl4)诱导的小鼠急性肝损伤的保护作用。方法:将60只昆明小鼠按体质量随机分为6组,即正常组,模型组,商陆总皂苷低、中、高剂量(25、50、100mg/kg)组,水飞蓟素(100mg/kg)组。每天灌胃给药1次,连续给药7d,末次给药1h后,采用腹腔注射CCl4诱导建立小鼠急性肝损伤模型,测定商陆总皂苷对血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)水平的影响;测定肝匀浆中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性以及丙二醛(MDA)含量变化;HE染色观察肝脏病理改变;免疫组化法检测Bax和Bcl-2表达。结果:与模型组比较,不同剂量商陆总皂苷均能显著降低血清中ALT、AST水平(P<0.01)。商陆总皂苷低剂量组肝匀浆CAT、GSH-Px活性显著升高(P<0.05,P<0.01),商陆总皂苷中、高剂量组肝匀浆SOD、CAT、GSH-Px活性显著升高(P<0.01),MDA含量显著降低(P<0.05,P<0.01);免疫组化结果显示商陆总皂苷能够减少促凋亡蛋白Bax的表达(P<0.01),增加抗凋亡基因Bcl-2的表达(P<0.01)。结论:商陆总皂苷对CCl4诱导的小鼠急性肝损伤具有较好的保护作用。
【Abstract】 Objective:To investigate the protective effect of total saponins from Phytolacca acinosa(SPA) on acute liver injury in mice induced by carbon tetrachloride(CCl4),and to explore the correlated mechanism.Methods:Sixty SPF KM mice were randomly divided into normal control group,model group,positive group,and SPA high-dose,medium-dose and low-dose(20,50,100 mg/kg) groups.The mice were pre-treated orally with SPA once daily for seven consecutive days.Acute liver injury model was induced by intraperitoneal injection of CCl4 at 1 hour after the last oral administration.The concentration of the serum ALT and AST were measured by extracting the eyeball blood.In order to substantiate whether SPA protects mice against CCl4-induced oxidative stress,MDA content and the activities of three antioxidative enzymes(GSH-Px,CAT and SOD) were measured.The pathological changes of liver were observed by HE staining and the expressions of Bax and Bcl-2 were measured by immunohistochemically staining.Results:Compared with the model group,the serum ALT and AST were remarkably decreased in all SPA treated groups(P<0.01).The activities of CAT and GSH-Px in liver homogenate were significantly increased in the low dose group of SPA(P<0.05,P<0.01).The activities of SOD,CAT and GSH-Px in the liver homogenate increased(P<0.01),and the content of MDA decreased significantly(P<0.05,P<0.01) in the middle and high dose groups of SPA.Further investigation demonstrated that pretreatment with SPA could markedly down-regulated the protein expression of Bax and up-regulated Bcl-2 expression(P<0.01).Conclusion:SPA has a significant protective effect against acute liver injury induced by CCl4 in mice.
