我国中低纬度地区白蜡DNA图谱构建及SSR标记遗传多样性分析
Construction of DNA Fingerprinting and Analysis of Genetic Diversity with SSR Markers for Fraxinus sp. in Mid-Latitudes and Low-Latitudes of China
【摘要】 白蜡是我国一种常见绿化树种,因纬度位置、海拔高度和海陆分布等自然地理因素不同,造成地区之间种质资源的自然差异。真实性和纯度是种质资源检测的重要指标之一,因此建立快速、精准、便捷的SSR分子标记技术对于白蜡种质的溯源及新品种授权具有重要意义。本研究以2013年山东省林业科学研究院在白蜡种质资源调查项目中采集并保存的38份白蜡为试材,利用SSR分子标记技术对我国不同纬度地区的白蜡种质资源进行指纹图谱构建,从150对候选引物中成功筛选出15条特异性强、条带清晰的引物作为核心引物,共检测出3.8个基因型,每对引物扩增的基因型2~6个不等。并构建白蜡DNA指纹图谱,采用5对引物组合即可将38份白蜡种质完全区分。聚类分析结果表明,白蜡种质间的亲缘关系与纬度因素具有一定的相关性。
【Abstract】 Fraxinus is a kind of common greening tree species in China.Because of the latitude,altitude,sea land distribution and other different natural geographic factors,the natural differences are present between regions and germplasm resources.The authenticity and purity are one of the important indexes for germplasm resource detection.Therefore,it is of great significance to establish a rapid,accurate and convenient DNA molecular marker technique for the traceability of Fraxinus germplasms and the authorization of new varieties.In this study,38 samples of Fraxinus collected and preserved by Shandong Academy of Forestry Sciences in 2013 were tested.The construction of DNA fingerprinting and analysis of genetic diversity with SSR markers were conducted for Fraxinus in mid-latitudes and low-latitudes.And 15 primers with high specificity and clear bands were selected as core primers from 150 pairs of candidate primers.A total of 3.8 genotypes were detected,and each pair of primers amplified 2~6 genotypes.The Fraxinus could be distinguished by 5 primers.This indicated that there were correlations between phylogenetic relationships and geographic origin of different varieties.
