大鼠学习记忆能力及其海马内TNF-α、IL-6含量的增龄性改变
The changes of learning and memory ability as well as hippocampal TNF-α and IL-6 levels in different age rats
【摘要】 目的探讨大鼠学习记忆能力及其海马内神经炎症因子肿瘤坏死因子α(tumor necrosis factor-alpha,TNF-α)及白细胞介素6(interleukin-6,IL-6)的增龄性改变。方法 SD大鼠42只,雌雄各半,分别为2月龄15只,6月龄13只,12月龄14只,Morris水迷宫检测不同月龄大鼠空间学习记忆能力,real time RT-PCR法检测大鼠海马TNF-α和IL-6 mRNA的表达,ELISA检测大鼠海马TNF-α和IL-6蛋白的含量。结果定位航行实验中,6月龄大鼠的平均逃避潜伏期最短,相比于12月龄,平均逃避潜伏期明显缩短(P<0.05),相比于2月龄,平均逃避潜伏期变化不明显(P>0.05),12月龄大鼠的学习成绩与2月龄比较无统计学意义(P>0.05);空间探索实验中,6月龄大鼠进入平台区次数最多、原平台象限滞留时间百分比及运动距离百分比最高,6月龄大鼠在原平台象限运动距离百分比较2月龄显著增加(P<0.05),而原平台象限滞留时间百分比及运动距离百分比变化不明显(P>0.05),6月龄大鼠进入平台区次数、原平台象限滞留时间百分比及原平台象限运动距离百分比与12月龄大鼠比较均显著增加(P<0.05),12月龄大鼠的学习成绩与2月龄大鼠比较差异无显著性(P>0.05)。real time RT-PCR结果表明,与2月龄及12月龄比较,6月龄大鼠海马TNF-α、IL-6 mRNA表达明显增高(P<0.01);ELISA结果显示,相比于2月龄及12月龄,6月龄大鼠海马TNF-α、IL-6蛋白含量均显著增高(P<0.01)。12月龄大鼠海马TNF-α、IL-6 mRNA及蛋白含量与2月龄比较无统计学意义(P>0.05)。结论大鼠学习记忆能力的增龄性改变与海马TNF-α、IL-6 mRNA及蛋白水平的增龄性改变呈对应关系。
【Abstract】 Objective To explore the changes of learning and memory ability and hippocampal tumor necrosis factor-alpha( TNF-α) and interleukin-6( IL-6) levels in different age rats. Methods 42 Sprague Dawley rats( male and female each half) were used: 2 months( n = 15),6 months( n = 13),and 12 months( n = 14).Learning and memory ability in different age rats were observed by Morris water maze,the levels of TNF-α,IL-6 mRNA and protein in the hippocampus were determined by real time RT-PCR and Western blot respectively.Results In the navigation experiment,the average escape latency in 6-month old rats was shortest,compared to 2-month rats,the change of average escape latency is not obvious( P > 0. 05),compared to 12-month rats,the average escape latency was significantly shortened( P < 0. 05),and there was no statistical significance between12-month and 2-month rats( P > 0. 05). In the space exploration experiment,6-month-old rats had longer times of passing platform,percent of residence and swim distance on original platform quadrant,compared with 2-month-old rats,the percent of swim distance on original platform quadrant in 6-month-old rats was significantly increased( P < 0. 05),while there were no significant changes in the times of passing platform and percentage of residence time on original platform quadrant in 6-month-old rats( P > 0. 05). Compared with 12-month-old rats,the time of passing platform,percent of residence time and swim distance on original platform quadrant in 6-month-old rats were increased obviously( P < 0. 05),and there is no significant difference in 12-month-old rats compared with 2-month-old rats( P > 0. 05). The results of real time RT-PCR showed that TNF-α and IL-6 mRNA were significantly increased in hippocampus of 6-month-old rats compared with 2 and 12 month-old-month rats( P < 0. 01). The results of ELISA showed that TNF-α and IL-6 protein levels were significantly increased in the hippocampus of 6-month-old rats compared with 2-and 12-month-old rats( P < 0. 01). There was no significant change in hippocampal TNF-α IL-6 mRNA and protein levels in 12-month-old rats compared with 2-month-old rats( P > 0. 05). Conclusion The changes of learning and memory ability in different age rats showed a corresponding relationship with hippocampal TNF-α,IL-6 mRNA and protein levels.
