NO及VEGF、sFlt-1在早发型子痫前期胎盘中的表达及意义

【摘要】 目的检测VEGF、sFlt-1及NO在早发型子痫前期和晚发型子痫前期患者血清和胎盘中的表达。方法随机选取2013年5月至2014年6月规律产检并分娩的子痫前期孕妇75例,其中早发型子痫前期31例,晚发型子痫前期44例。选取同期健康晚期妊娠孕妇30例作为健康对照组,采用免疫组织化学SP法检测VEGF、sFlt-1的表达,用硝酸还原法测定胎盘中NO的表达,并进行相关性分析。结果早发型子痫前期患者胎盘组织中VEGF阳性表达率32.2%,晚发型组为61.4%,正常对照组为86.7%,3组差异有统计学意义(P<0.05);早发型子痫前期患者胎盘组织中sFlt-1阳性表达率82.1%,晚发型组74.8%,正常对照组12.9%,3组差异有统计学意义(P<0.05);NO的光密度结果在早发型子痫前期患者胎盘组织中是59.3±2.21,晚发型组为31.6±1.90,正常对照组89.9±2.17,3组差异有统计学意义(P<0.05)。结论 sFlt-1在早发型子痫患者的胎盘组织中表达最高,抑制了其靶组织VEGF表达,影响了胎盘微血管形成;NO在早发型子痫患者的胎盘组织中表达最低,抑制了微血管的舒张。SFlt-1和NO与早发型子痫前期的发病密切相关。

【Abstract】 Objective:To explore the relationship between the genetic expression of VEGF,sFlt-1,NO in the placentas of patients with early onset preeclampsia and vessel diseases of placentas,compared with late onset ones. Methods:75 cases were randomly selected among all the patients with preeclampsia,who had come for regular antenatal care and delivery. Among those cases,31 were early onset while the rest ones were late onset. Another 30 cases of healthy pregnant women were chosen as the control group. The immunohistochemistry SP method is utilized to detect the expression of VEGF,and sFlt-1. Additionally,The expression of NO in placenta was determined by silver nitrate reduction method and furtherly the relativity analysis. Results:For the expression rate of VEGF in early onset preeclampsia cases,inside the placenta tissues,it is 32.2%;that in the late onset cases is 61.4%,and 86.7% for the control group. The difference is meaningful statistically(P<0.05). For the expression rate of sFlt-1,it is 83.9% for early onset cases,77.3% for late onset cases,and 13.3% for control ones. The difference is statistically meaningful(P<0.05). The optical density results of NO in early preeclampsia is 59.3±2.21;that in the late onset cases is31.6±1.90,and 89.9±2.17 for the control group. The difference is meaningful statistically(P<0.05).Conclusion:sFlt-1 is highly expressed in the early onset preeclampsia patients,which suppresses that of VEGF inside its target tissue,affecting the formation of capillaries in placenta tissue and leading to the preeclampsia. NO in the early_onset preeclampsia expression of the lowest,inhibiting vascular relaxation. sFlt-1 and NO is closely related to early onset of preeclampsia
