蒺藜皂苷对人黑素瘤A375细胞酪氨酸酶和小眼相关转录因子表达的影响
Effect of Thistle Saponins on TYR and MITF mRNA Expressions of A375 Cells
【摘要】 目的:在前期研究的基础上,以蒺藜皂苷为研究对象,考察其对A375细胞酪氨酸酶活性、TYR和MITF mRNA的表达的影响。方法:采用不同浓度的蒺藜皂苷作用于A375细胞48 h后,应用左旋多巴氧化法测定酪氨酸酶活性,荧光定量PCR法检测A375细胞TYR和MITF mRNA的表达情况。结果:应用蒺藜皂苷处理后,酪氨酸酶活性均有不同程度的增加,当达到100μg/m L和200μg/m L时,有统计学差异。TYR和MITF mRNA的表达明显增强(P<0.05)。结论:蒺藜皂苷可能通过增强A375细胞TYR和MITF mRNA的表达,以提高酪氨酸酶活性,促进黑素合成,从而为蒺藜皂苷的开发与应用奠定基础。
【Abstract】 Objective: On the basis of the previous research,regarding thistle saponins as the research object,to study its tyrosinase activity,tyrosinase and MITF mRNA expressions of A375 cells. Methods: Different concentrations of thistle saponins were used for the A375 cells for 48 hours,and then the tyrosinase activity was evaluated by levodopa oxidation method. TYR and MITF mRNA expressions of A375 cells were detected by luorescence quantitative PCR. Results: After application of thistle saponins,the activity of tyrosine was increased to a certain degree. When it was up to 100 μg / mL and 200 μg / mL,there was a statistical significance. TYR and MITF mRNA expressions were obviously increased( P < 0. 05). Conclusion: Thistle saponins can improve the activity of tyrosinase by enhancing TYR and MITF mRNA expressions of A375 cell,so as to promote the synthesis of melanin,which lay a foundation for the development and application of thistle saponins.
