人参皂苷Rb1对川崎病小鼠治疗作用的研究
Effect of Ginsenoside Rb1 on Kawasaki Disease in Mice
【摘要】 目的:用干酪乳杆菌细胞壁提取物(LCWE)诱导川崎病(KD)小鼠模型,探讨人参皂苷Rb1治疗川崎病小鼠的作用。方法:LCWE组小鼠每只腹腔注射LCWE 0.5 mL(1 mg/mL),Rb1组给予20 mg/kg和40 mg/kg Rb1治疗,ASP组灌胃给予250 mg/kg ASP治疗。观察实验期间小鼠的变化,对小鼠进行眼眶取血并解剖心、脾、肾、胸腺等脏器进行病理切片,以及对小鼠血液进行血常规检验和测定血清中的肿瘤坏死因子(TNF-a)的含量。结果:小鼠体质量明显降低,血常规检测LCWE造模组白细胞(WBC)含量明显升高,Rb1组下降。血清中肿瘤坏死因子(TNF-a)含量明显升高,Rb140 mg/kg组下降。病理切片显示LCWE造模组冠状动脉血管壁增厚并伴有炎性浸润,Rb1组血管壁变薄炎性浸润减少。结论:人参皂苷Rb1对川崎病小鼠有一定的治疗作用。
【Abstract】 Objective: To explore the effect of Ginsenoside Rb1 on Kawasaki disease( KD) in mice. Methods: Lactobacillus casei cell wall extract( LCWE,1 mg/mL,ip) was used to induce KD model. The mice were randomly divided into the control group,the model group,Rb1 groups( 20 mg/kg,40 mg/kg),and ASP group( 250 mg/kg). Then the changes of mice during the experiment were observed. Orbital blood samples were collected and the tissue samples of the heart,spleen,kidney and thymus were collected for HE staining,and TNF-a level was measured. Results: Body weight of the mice had decreased,WBC levels increased significantly in the model group,compared to the control group,but Rb1 inhibited the effect of LCWE. TNF-a level was significantly increased after modeling,but it was decreased in Rb1 group( 40 mg/kg). Pathological measure showed that the coronary artery wall was thickened with serious inflammatory infiltration in the model group,vascular wall became thinner with less inflammatory infiltration in Rb1 groups. Conclusion: Ginsenoside Rb1 has a certain therapeutic effect on KD in mice.
