黄芪提取物对老年痴呆大鼠NF-kB信号通路相关蛋白表达的影响

【摘要】 目的研究黄芪提取物(EA)对老年痴呆(AD)大鼠NF-k B信号通路相关蛋白的影响。方法大鼠海马内注射Aβ25-35制备AD大鼠模型,实验分4组:对照组、模型组、黄芪组和西药治疗组。黄芪组和西药治疗组分别用黄芪提取物和脑复康灌胃,对照组和模型组用等量生理盐水灌胃。用Morris水迷宫测定各组大鼠的学习记忆能力,Western blot测定各组大鼠海马NF-k B、Ik B-a和caspase-3蛋白表达水平。结果模型组的逃避潜伏期延长,单位时间内跨越原平台次数减少,NF-k B和caspase-3蛋白表达升高,而Ik B-a蛋白表达降低,与对照组比较差异均有统计学意义(均P<0.05);黄芪组的逃避潜伏期缩短,单位时间内跨越原平台次数增多,NF-k B和caspase-3蛋白表达降低,而Ik B-a蛋白表达升高,与模型组比较差异均有统计学意义(均P<0.05),其作用效果与西药治疗组相当(P>0.05)。结论黄芪提取物通过提高AD大鼠海马区Ik B-a的含量,使游离NF-k B的含量减少,激活caspase-3受到抑制,从而抑制了神经细胞的凋亡。

【Abstract】 [Objective]To study the effects of astragalus extract on related protein expression of NF-k B signal pathway in AD rats.[Methods]The AD model rats were established by injecting Aβ 25-35 into districts of hippocampuses. The experiment was divided into 4 groups:control group,model group,astragalus group and western medicine treatment group. Astragalus and western medicine treatment group were treated with astragalus extract and piracetam by gavage respectively,and control and model group were treated with isodose physiological saline by gavage. The ability of learning and memory was detected by Morris water maze,and the protein expression levels of NF-k B、Ik B-a and caspase-3 in the hippocampus of each group rats were detected by western blot.[Results]The incubation period of avoiding lengthened,the frequency of passing through platform decreased and the expression levels of NF-k B and caspase-3 increased in model group rats,while the expression level of Ik B-a decreased,compared with the control group,the differences were statistically significant(all P <0.05). The incubation period of avoiding obviously shortened,the frequency of passing through platform increased and the expression levels of NF-k B and caspase-3 decreased,while the expression level of Ik B-a increased in astragalus group rats,compared with the model group,the differences were statistical significant(all P<0.05). Its effect was similar with the western medicine treatment group(P>0.05).[Conclusion]The astragalus extract can inhibit the apoptosis of nerve cells through increasing the content of Ik B-a and decreasing the content of free NF-k B in the Hippocampus of AD rats to restrain the activation of caspase-3.
