土家族麝针疗法“活血生新”治疗缺血性脑中风的作用机制研究
Study on Mechanism of Activating Blood and Creating New Blood Vessel by Musk Needle Therapy in Tujia Medicine on Ischemic Stroke Rat Model
【摘要】 目的:探讨土家族麝针疗法治疗缺血性脑中风的作用机制,揭示其活血化瘀诱导血管新生的药理作用。方法:采用改良线栓法建立缺血性脑中风大鼠模型,以Zea-longa为1~3分的大鼠为研究对象,以土家族麝针疗法干预3个疗程后,TTC染色法观察其对大脑梗死灶体积的影响,激光散斑成像技术观察其对脑皮质区整体血流速度的影响,western-blot法测定血管内皮生长因子(VEGF)及其受体1(VEGFR1)的表达水平。结果:与正常组比较,缺血性脑中风大鼠脑梗死体积显著增加,脑皮质区血流速度明显减缓,脑组织中VEGF蛋白和VEGFR1蛋白表达水平均显著增加;经给予土家族麝针疗法治疗后,脑梗死体积显著降低,脑皮质区的血流速度显著加快,脑组织中VEGF蛋白和VEGFR1蛋白表达水平显著增加,且其影响随着疗程的延长更为显著。结论:土家族麝针疗法能显著改善缺血性脑中风所致的脑梗死体积,其"活血生新"的作用机制可能与改善脑皮质区的血流速度、增加血管内皮细胞生长因子及其受体表达水平有关。
【Abstract】 Objective: To explore the mechanism of activating blood and creating new blood vessel by musk needle therapy in Tujia medicine on ischemic stroke rat model. Methods: Ischemic stroke rat model was established by reforming Longa method. The ischemic stroke rats with the Zea-longa score 1 to 3 were divided into model group and treatment group,the latter was treated by musk needle therapy for 3 courses of treatment while the former without any further processing,the normal rats were used as the control group. Tetrazolium method was used to observe the volume of cerebral infarction,laser speckle imaging was used to observe the blood flow rate of the whole cerebral cortex and western-blot was used to detect the expression level of vascular endothelial growth factor( short for VEGF) and VEGF receptor 1. Result: Comparing with the control group,the volume of cerebral infarction,the expression level of VEGF and VEGF receptor 1 increased significantly while the blood flow rate of the whole cerebral cortex decreased significantly both in model group and treatment group. After given musk needle therapy,the volume of cerebral infarction decreased significantly while the blood flow rate of the whole cerebral cortex,the expression level of VEGF and VEGF receptor 1 increased significantly,and these results have a positive relationship with the courses of treatment. Conclusions: Musk needle therapy in Tujia medicine could significantly improve the volume of cerebral infarction caused by ischemic stroke,its mechanism of activating blood and creating new blood vessel may have something to do with its functions on increasing the blood flow rate of the whole cerebral cortex,the expression level of VEGF and VEGF receptor 1.
