【摘要】 目的:探讨峨参内酯与5-FU联用对人结肠癌SW480三维立体多细胞球模型的作用及其机制。方法:采用CCK8法检测峨参内酯及峨参内酯与5-FU联用对SW480三维立体多细胞球模型的抑制作用;采用流式细胞仪观察分析峨参内酯与5-FU联用对SW480三维立体多细胞球模型细胞周期和细胞凋亡的影响;采用蛋白质印迹(Western blot)技术检测分析峨参内酯与5-FU联用对SW480三维立体多细胞球模型细胞中FAK、COX2蛋白表达的影响。结论:峨参内酯对人结肠癌SW480三维立体多细胞球模型有明显的抑制作用,呈现出一定的药物剂量依赖性,其半数致死量分别为26.82μg/m L;峨参内酯与5-FU联用时,细胞活力显著降低,且随着峨参内酯浓度的加大而逐渐降低;5-FU可使SW480细胞周期停止在S期,峨参内酯可使SW480细胞周期停止在S期、G2/M期,两种药物配合使用,可使SW480细胞周期停止在S期、GA/M期并最终诱导细胞凋亡,并且能够明显抑制细胞中FAK、COX2两种蛋白的表达。更多还原

【Abstract】 Objective: To investigate the effects of Anthricin combined with fluorouracil and the mechanism on SW480 three-multicellular spheroids models,providing the basic research data for further elucidate the anti-tumor mechanism of Anthriscus sylvestris.Methods: We established the SW480 three-dimensional multi-cellular spheroids model and tested Anthricin combined with fluorouracil in the inhibition effects on SW480 three-dimensional multicellular spheroids models by CCK8 method.We tested Anthricin combined with fluorouracil in the influnences of the cell cycle and apoptosis on SW480 three-dimensional multicellular spheroids models by the flow cytometry and detected the influences of Anthricin combined with fluorouracil in expressions of FAK and COX2 protein in SW480 three-dimensional multicellular spheroids models.Results: Anthricin could obviously inhibit SW480 three-dimensional multicellular spheroids models and it showed a certain dose-dependent.The half lethal dose was 26.82 μg/m L.Anthricin combined with fluorouracil could significantly decrease the activity of SW480 three-dimensional multicellular spheroids models and the activity would decrease with the Anthricin concentration increase.5-FU could make SW480 cells’ cycle be arrested in S phase and Anthricin could make their cell cycle be arrested in S phase and G2/M phase.Anthricin combined with fluorouracil could make SW480 cells’ cycle be arrested in S phase and G2/M phase and ultimately induce cell apoptosis,and they could obviously inhibit the expressions of FAK and COX2 in SW480 cells.更多还原
