金钗石斛多糖对肝纤维化大鼠转化生长因子及基质金属蛋白酶的影响
influence of TGF and MMP in Dendrobium Nobile Polyose on Hepatic Fibrosis Rats
【摘要】 目的:探讨金钗石斛多糖对肝纤维化大鼠转化生长因子β1(TGFβ1)、基质金属蛋白酶抑制因子(TIMP-1)及基质金属蛋白酶(MMP13)的影响。方法:SD大鼠按随机数字表法分为正常对照组,模型组,秋水仙碱组,扶正化瘀组,石斛多糖低、中、高剂量组,采用四氯化碳(CCL4)复合乙醇诱导大鼠肝纤维化模型,造模8周成功后,分别给予相应的干预药物,正常组、模型组给予生理盐水,每日1次,连续4周。采用荧光定量PCR技术检测大鼠肝脏TGFβ1、TIMP-1、MMP13 mRNA的表达差异。结果:与模型组相比,各治疗组肝组织内纤维化程度均有不同程度减轻;与正常对照组比较,模型组TGFβ1、TIMP-1 mRNA的表达上调(P<0.05),MMP13 mRNA的表达下调(P<0.05);与模型组比较,各药物干预组均能降低大鼠肝组织TGFβ1、TIMP-1 mRNA的表达,其中扶正化瘀组、石斛多糖中、大剂量表达有显著性差异(P<0.05);与模型组比较,各药物干预组提高MMP13 mRNA的表达,其中以扶正化瘀组、石斛多糖大剂量为甚(P<0.05)。结论:石斛多糖具有下调TGFβ1、TIMP-1 mRNA表达,上调MMP13 mRNA表达,可能是其抗肝纤维化的作用机理之一。
【Abstract】 Objective:To study the influence of transforming growth factor-β1( TGF-β1),tissue inhibitor of metalloproteinases-1( TIMP-1) and matrix metalloproteinase( MMP-13) in dendrobium nobile polyose on hepatic fibrosis rats.Methods:SD rats were randomly divided into normal group,model group,colchicine group,Fuzheng Huayu group,caulis dendrobii low-dose group,caulis dendrobii medial-dose group,caulis dendrobii high-dose group.SD rat models of hepatic fibrosis were induced by carbon tetrachloride composite ethanol( CCl4).After 8 weeks,each group was given corresponding drug intervention respectively.The normal group and model group were given normal saline once a day for 4 weeks.Use fluorescence quantitative PCR technique to detect mRNA expression differences of TGF-β1,TIMP-1and MMP-13 in liver.Results:Compared with normal group,the mRNA up-regulated expressions of TGF-β1 and TIMP-1( P < 0.05) and the mRNA down-regulated expression of MMP-13( P < 0.05) in model group.Compared with model group,the each drug intervention group can reduce mRNA expressions of TGF-β1 and TIMP-1.The expressions in Fuzheng Huayu group,caulis dendrobii medial-dose group and caulis dendrobii high-dose group had significant differences( P < 0.05).Compared with model group,each drug intervention group can improve mRNA expression of MMP-13,especially in Fuzheng Huayu group and caulis dendrobii high-dose group( P < 0.05).Conclusion:The dendrobium nobile polyose can reduce mRNA expressions of TGF-β1 and TIMP-1 and improve mRNA expression of MMP-13.It may be one of anti-hepatic fibrosis mechanisms.
