潜阳育阴颗粒对SHR大鼠肾脏CTGF的影响
Effect of Qianyang Yuyin Granules on Kidney CTGF of SHR Rats
【摘要】 目的:研究潜阳育阴颗粒对高血压肾病的治疗作用与机制。方法:随机将32只12周龄雄性SHR大鼠等分为4组:模型组(SHR)、潜阳育阴组(SHR+Q)、贝那普利组(SHR+B)、及潜阳育阴+贝那普利组(SHR+Q+B)。另取8只年龄、性别配对的WKY大鼠为正常组。SHR+Q、SHR+B及SHR+Q+B组分别以潜阳育阴混悬液5 g/(kg·d)、贝那普利混悬液1.67 mg/(kg·d)及潜阳育阴混悬液5 g/(kg·d)+贝那普利混悬液1.67mg/(kg·d)灌胃。正常组及模型组不给药,予等量蒸馏水灌胃,五组均正常饮食。灌胃周期共8周。实验前及实验后均采用无创伤性尾静脉测压法测量大鼠血压。实验8周后,用大鼠代谢笼按组分别装大鼠,接取各组大鼠24 h的尿液,采用全自动生化仪和放免法检测大鼠尿m-ALB及尿NAG含量;HE染色观测肾脏病变;Masson染色观察肾组织纤维化;Elisa法检测肾组织Pro、Ald、Ang II含量;Western Blot法检测肾组织ACE、AT1R、CTGF蛋白表达。结果:SHR组血压显著增加,尿液中m-ALB、NAG浓度增高,潜阳育阴颗粒可以降低血压与m-ALB、NAG。模型组出现Ang II、AT1R、CTGF活性的上调,而潜阳育阴颗粒对其有抑制作用。结论:潜阳育阴药物对高血压肾病有治疗作用,减轻高血压肾病过程中的纤维化,推测其治疗高血压肾脏病可能与其干预CTGF通路有关。
【Abstract】 Objective: To study the effect of Qianyang Yuyin Granules on the hypertensive nephropathy and its mechanism. Methods: Thirty-two male SHR rats,12 weeks old,were randomly divided into four groups: model group(SHR),Qianyang Yuyin Granules group(SHR + Q),Benazepril group(SHR B) and Qianyang Yuyin Granules + Benazepril group group(SHR + Q + B). Another eight WKY rats which had the matched age and gender were as the normal group.Blood pressure of rats was measured before and after the experiment. By 8 weeks treatment,24 h urine of rats was collected and the rats were put into the metabolism cages. M-ALB and NAG in urine were tested with automatic biochemistry analyze. HE dyeing was used for observing kidney disease(CKD) and Masson staining for observing the renal tissue fibrosis. Pro,Ald and Ang II levels in kidney were detected by ELISA method. ACE,AT1 R and CTGF protein expressions were detected by Western Blot method. Results: The blood pressure of SHR groups was increased significantly while m-ALB and NAG in urine increased. Qianyang Yuyin Granules can reduce blood pressure,m-ALB and NAG. Ang II,AT1 R and CTGF activity in model group increased and Qianyang Yuyin Granules inhibited their activity. Conclusion:Qianyang Yuyin Granules have the good effect on hypertensive nephropathy and there is some relation with CTGF pathways.
