福州2009-2014年甲型H1N1流感病毒株HA基因进化分析
Evolution Anaysis of HA Genome Sequence Analysis in Fuzhou from 2009 to 2014
【摘要】 目的:分析福州地区2009-2014年甲型H1N1流感病毒株的HA基因序列和遗传特征。方法:采集流感样病例的咽拭子标本,用MDCK细胞培养法和Real-time RT-PCR法对6株分别来自2009-2014年的病毒株进行病毒分离、鉴定,HA基因片段通过RT-PCR法进行扩增后克隆到p MD18-T载体上,并测定6株病毒株的HA基因序列。通过生物软件Bio Edit对该分离株和Gen Bank中相关毒株的序列进行基因进化树分析。结果:6株分离株的HA基因推导氨基酸序列与参考株A/California/04/2009相比,2009-2014年HA1区氨基酸发生了突变,主要是位于136、145、188、189、192和193位点。结论:近几年,福州地区H1N1流感的HA基因序列与世界流行的病毒参考株具有高度同源性,并多处氨基酸发生替换而产生抗原性漂移。
【Abstract】 Objective:To investigate HA genetic characteristic of influenza type A H1N1 virus in Fuzhou from 2009 to 2014.Method:Acquisition of influenza like cases of throat swab specimens by MDCK cell culture method and Real-time RT-PCR method in 6 strains from 2009 to 2014 were isolated and identified. The HA gene fragment was amplified and cloned into p MD18-T vector by RT-PCR method, and determination of HA gene sequences of 6 virus strains.The phylogenetic tree of the isolates and Gen Bank was analyzed by Bio Edit.Result:The deduced amino acid sequence of HA gene of 6 strains compared to reference virus A/California/04/2009, HA protein of the isolate viruses from 2009 to 2014 had amino acid mutation in sites of 136,145,188,192 and 193.Conclusion:In recent years,the HA gene sequence of H1N1 influenza in Fuzhou region is highly homologous to the world’s most popular virus reference strains,and many amino acids are replaced to produce antigenic drift.
