NMR法测定A,C,Y和W135群脑膜炎球菌荚膜多糖氧乙酰基含量

【摘要】 目的:采用核磁共振(nuclear magnetic resonance,NMR)技术检测A,C,Y和W135群脑膜炎球菌荚膜多糖(groups A,C,Y,W135 Neisseria meningococcal capsular polysaccharides,GAMP,GCMP,GYMP,GWMP)氧乙酰基含量。方法:对A,C,Y和W135群脑膜炎球菌荚膜多糖样品进行溶解,离心,转移等处理步骤,采用核磁共振谱仪检测,并对检测参数D1,NS和计算标准进行优化使其满足定量检测要求。结果:确定的检测条件:D1值GAMP为18,GCMP,GYMP和GWMP为20;NS值均为128次,计算标准GAMP采用H-3和H-4与H1积分面积之比,GCMP采用H-7和H-8与H-3e积分面积之比,GYMP和GWMP采用H-7和H-9与H-3e积分面积之比。结论:NMR法简便、快速、准确,可用于检测A,C,Y和W135群脑膜炎球菌荚膜多糖氧乙酰基含量。

【Abstract】 Objective: To determine the O-acetyl content of groups A,C,Y and W135 Neisseria meningococcal capsular polysaccharides( GAMP,GCMP,GYMP,GWMP) by nuclear magnetic resonance( NMR) method.Methods: After being processed with dissolving,centrifuging and transferring,the sample of Neisseria meningococcal capsular polysaccharides was monitored by NMR,and the test parameters such as D1,NS and calculating standards were optimized to meet the requirements of quantitative determination.Results: The detecting condition was determined to be as follows: the D1 values of GAMP was 18,and those of GCMP,GYMP and GWMP were all 20;the NS values of GAMP,GCMP,GYMP and GWMP were all 128; the calculating standard of GAMP was comparing the ratios of H-3 and H-4 proton signals with H1 signal,and that of GCMP was comparing the ratios of H-7 and H-8 proton signals with H-3 e signal.The calculating standards for both of GYMP and GWMP were comparing the ratios of H-7 and H-9 proton signals with H-3 e signal.Conclusion: The NMR method is simple,rapid and accurate.It is suitable for O-acetyl content determination of GAMP,GCMP,GYMP and GWMP.
