不同保存时间的红细胞携氧能力和体内存活变化研究

【摘要】 目的探讨血液保存时间对红细胞携氧能力及体内存活率的影响。方法检测不同保存期血样pH值、Na~+、K~+、葡萄糖、乳酸盐、乳酸脱氢酶、三磷酸腺苷(ATP)和2,3-二磷酸甘油酸(2,3-DPG)的浓度。采用血氧分析仪和流式细胞仪检测血样携氧能力和体内存活率。结果红细胞悬液的乳酸盐、乳酸脱氢酶和K~+浓度随库存时间增加而增加,悬液p H值、Na~+、葡萄糖、ATP和2,3-DPG浓度随库存时间增加而降低。有效携氧量(Q)、P50和体内存活率存时间增加而减少,并都与库存时间呈线性关系(r=-0.985、r=-0.989、r=-0.985,P<0.01);Q与P50、Q与体内存活率和P50与体内存活率也呈线性关系。结论体内存活率与Q和P50紧密相关,依据Q值和P50计算不同库存时间红细胞体内存活率。Q相同的红细胞悬液在体内有几乎相同的存活红细胞数。

【Abstract】 Objective To evaluate the effects of storage time on oxygen-carrying capacity and survival of red blood cells( RBCs). Methods The p H value and the concentrations of Na~+,K~+,glucose,lactate,lactic dehydrogenase,adenosine triphosphate( ATP) and 2,3-diphosphoglyceric acid( 2,3-DPG) in RBC suspensions with different storage periods were detected. Oxygen-carrying capacity and survival of RBCs with different storage periods were determined by oxygen gas analyzer and flow cytometry,respectively. Results The concentrations of lactate,lactic dehydrogenase and K~+in RBC suspension increased with storage time increasing,while the p H value and the concentrations of Na~+,glucose,ATP and 2,3-DPG in RBC suspension decreased with storage time increasing. Effective oxygen-carrying amount( Q),P50 and survival of RBCs all decreased with storage time increasing,and they were all linear to storage time( r =-0. 985,r =-0. 989,r =-0. 985,P <0. 01). Furthermore,Q and P50 were both linear to survival,and Q was also linear to P50. Conclusion There exists a very close relation between survival and Q and P50,and the survival rate can be figured out when Q or P50 is known. The RBC suspensions with same Q have almost the same account of survival RBC.
