三七皂苷S-6的溶血性及体外免疫活性研究

【摘要】 研究三七皂苷S-6的溶血性及体外免疫活性。常规方法提取PNS,大孔树脂柱层析分离纯化得S-6。溶血试验测定红细胞溶血率。脾淋巴细胞毒性试验确定S-6的安全剂量范围,体外脾淋巴细胞增殖反应和IL-2的诱生及含量测定检测S-6的体外免疫活性。S-6溶血性较小,2 mg/mL浓度以下无溶血性;S-6(0.1μg/mL、1μg/mL)显著促进了ConA诱导的小鼠脾淋巴细胞增殖反应,且与PNS和APS组相比,差异不显著;S-6(1μg/mL)极显著地促进了LPS诱导的小鼠脾淋巴细胞增殖反应,且与PNS相比,S-6(1μg/mL)差异显著,其余各个剂量与PNS和APS组相比,差异不显著;S-6(12.5μg/mL、25μg/mL、50μg/mL)显著或极显著地提高了IL-2的含量,且与PNS和APS组相比,差异不显著。S-6安全性好,体外具有较好的细胞免疫活性和诱生细胞因子的能力。

【Abstract】 To investigate the hemolytic characteristics and biologic immunocompetence of S-6 in vitro, PNS was extracted using butyl alcohol and S-6 was obtained by separation and purification with D101 macroreticular resin. The in vitro hemolytic activity, range of safe dose and immunocompetence of S-6 were examined for parameters including hemolysis, inducible and content determination of IL-2, splenocyte toxicity and proliferation. The hemolytic activity of S-6 was lower than 2 mg/mL. S-6 significantly increased concanavalin A-stimulated splenocyte proliferation in vitro at 0.1 μg/mL. However, S-6 had no significant difference with PNS and APS at 1 μg/mL. S-6 significantly increased concanavalin lipopolysaccharide-stimulated splenocyte proliferation in vitro at 1 μg/mL and had significant difference with PNS. S-6 also increased the content of IL-2 at 12.5 μg/mL, 25 μg/mL and 50 μg/mL and had no significant difference with PNS and APS. In conclusion, S-6 had high security and increased cellular response and in vitro content of IL-2.
