【摘要】 目的:探讨威灵仙水提物对阿霉素诱导耐药的人甲状腺未分化癌细胞株HTh74Rdox增殖的影响及机制。方法:用不同浓度威灵仙水提物处理HTh74Rdox后观察细胞形态;噻唑蓝(MTT)比色法检测威灵仙水提物对HTh74Rdox细胞的增殖抑制作用;流式细胞术检测HTh74Rdox细胞凋亡;蛋白质免疫印迹法(Western blot)检测凋亡相关半胱氨酸蛋白酶-3(Caspase-3)及其活性片段(cleaved-Caspase-3),B淋巴细胞瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)的表达。结果:威灵仙水提物对HTh74Rdox细胞的增殖有明显抑制作用,联合阿霉素后可减少阿霉素剂量;威灵仙水提物可显著诱导细胞凋亡(P<0.01);且能明显提高cleaved-Caspase-3/Caspase-3,Bax/Bcl-2(P<0.05)。结论:威灵仙水提物可以抑制HTh74Rdox细胞的增殖,其机制可能与提高cleaved-Caspase-3/Caspase-3,Bax/Bcl-2有关;联合化疗药物使用具有减毒增效的作用。更多还原

【Abstract】 Objective: To explore the effect of water extract of Clematidis Radix et Rhizoma on proliferation of HTh74 Rdox cells and its mechanism. Method: HTh74 Rdox cells were treated by different concentrations of water extract of Clematidis Radix et Rhizoma,in order to observe morphological changes and apoptosis of HTh74 Rdox cells. methylthiazolyldiphenyl-tetrazolium bromide(MTT) was used to test the inhibitory effect of water extract of Clematidis Radix et Rhizoma on proliferation of HTh74 Rdox cells. The cell cycle change and the cell apoptosis rate were further detected by flow cytometry; Western blot was used to measure the expressions of cleaved-Caspase-3,Caspase-3,Bcl-2 and Bax. Result: Zedoary oil played a significant role in inhibiting the growth of HTh74 Rdox cells. The combination of water extract of Clematidis Radix et Rhizoma and adriamycin could reduce the average dose of Adriamycin. Water extract of Clematidis Radix et Rhizoma significantly increased the percentage of cleaved-Caspase-3/Caspase-3 and Bax/Bcl-2. Conclusion: Water extract of Clematidis Radix et Rhizoma can inhibit the growth of HTh74 Rdox cells. And its possible mechanism may be correlated with the increase of cleaved-Caspase-3/Caspase-3 and Bax/Bcl-2 ratio. Water extract of Clematidis Radix et Rhizoma combined with chemotherapy drug has the effect in reducing toxicity and increasing efficiency.更多还原
