miR-10a-5p抑制人食管鳞状细胞癌的迁移和侵袭能力

【摘要】 目的探讨miR-10a在食管鳞状细胞癌(ESCC)中的表达水平及临床意义。方法实时荧光定量PCR技术(qRT-PCR)检测48例ESCC患者肿瘤组织及瘤旁正常组织中miR-10a的表达水平,同时检测相同患者术前血清及25例健康志愿者血清中miR-10a的表达水平,并分析其与临床特征之间的关系。另外,探索miR-10a对ESCC细胞迁移及侵袭能力的影响。结果 miR-10a在食管癌组织及术前患者血清中的表达水平要低于瘤旁正常组织及健康志愿者,miR-10a在肿瘤组织中的表达水平与淋巴结转移、TNM分期密切相关(P<0.05),miR-10a在术前患者血清中的表达水平与淋巴结转移、TNM分期、肿瘤分化程度密切相关(P<0.05)。miR-10a的过表达可以抑制细胞增殖及迁移、侵袭能力。结论作为一种抑癌基因,miR-10a成为ESCC诊断及治疗的潜在靶点。

【Abstract】 Objective To investigate the expression and clinical significance of miR-10 a in esophageal squamous cell carcinoma(ESCC).Methods Real time quantitative PCR(qRT-PCR) was used to detect the expression of miR-10 a in ESCC tissues and adjacent normal tissues of 48 patients.At the same time,the expression level of miR-10 a in serum of the same patients and 25 healthy volunteers were detected.The relationship between the miR-10 a expression and the clinical features was analyzed.In addition,we explored the effect of miR-10 a on the migration and invasion of ESCC cells.Results We found that miR-10 a expression levels in tumor tissues and preoperative serum of esophageal carcinoma patients were lower than that of non-tumorous tissues and healthy volunteers.MiR-10 a expression in tissues was closely related to lymph node metastasis and TNM stage(P<0.05),and miR-10 a expression in serum correlated to lymph node metastasis,TNM stage and tumor differentiation(P<0.05).Overexpression of miR-10 a could inhibit cell proliferation,migration and invasion.Conclusion As a tumor suppressor gene,the expression level of miR-10 a would be a potential biomarker for diagnosis and treatment of ESCC.
