钙离子对小鼠肝脏铁调素表达的影响及其调节机制

【摘要】 目的探讨钙离子对小鼠肝脏铁调素(hepcidin)mRNA表达的影响及其机制。方法实验小鼠随机分3组,对照组腹腔注射0.9%生理盐水,实验组腹腔注射CaCl2溶液(140 mg/kg),注射0、6、24 h后分别取材。试剂盒检测血清铁和转铁蛋白饱和度,RT-PCR检测肝脏hepcidin mRNA表达,Western印迹检测脾脏Fpn1蛋白和肝脏中几个关键信号分子的表达。结果相对于对照组,小鼠腹腔注射CaCl2溶液6、24 h后血清铁明显下降(P<0.05),肝脏中hepcidin mRNA表达在6 h下降显著(P<0.05),Fpn1蛋白表达上升,p-SMAD蛋白在6 h略有下降,而p-STAT蛋白在6 h后下降明显。结论 SMAD、STAT信号分子参与了钙离子对小鼠肝脏hepcidin mRNA的表达。

【Abstract】 Objective To study the effect and mechanism of calcium on the expression of m RNA hepcidin in the liver of mice.Methods The experimental mice were randomly divided into 3 groups,the control group was injected with 0.9% normal saline,the experimental group was injected with CaCl2 solution( 140 mg / kg),0 h,6 h and 24 h respectively. Detection of serum iron and transferrin saturation,RT-PCR was performed for m RNA of hepcidin. Fpn1 protein and signaling molecules were determined by Western blot. Results Compared with those of control group,calcium ion reduced the serum iron and m RNA hepcidin expressions significantly( P < 0. 05),increased Fpn1,p-SMAD and p-STAT protein expressions.Conclusions Calcium ion could reduce hepcidin expression and affect iron metabolism. The SMAD and STAT pathway is involved in the regulation of hepcidin.
