均匀设计法筛选淡竹叶内碳苷黄酮优化配比的实验研究
Uniform designed research on the optimum proportion of C-glycosylflavones fromLophatherum gracile
【摘要】 为优化淡竹叶内主要碳苷黄酮的配比,利用均匀设计法,选择U12×(107)表设计实验方案,以淡竹叶内主要的7种碳苷黄酮为考察因素,采用氧糖剥夺/复氧(oxygen glucose deprivation/reoxygenation,OGD/R)诱导心肌细胞的损伤模型,以噻唑蓝(MTT)法检测的细胞活力作为考察指标,优化碳苷黄酮配比并进行验证。结果表明:优化后的淡竹叶碳苷黄酮质量浓度配比为:异荭草苷∶日当药黄素∶阿福豆苷∶木犀草苷=0.822∶0.114∶0.268∶1.001,其细胞存活率显著上升(P<0.01)。基于均匀设计法优化的淡竹叶内碳苷黄酮配比可靠,具有很强的保护心肌细胞的作用。
【Abstract】 To optimize the proportion of main C-glycosylflavones fromLophatherum gracile,by employing U12×(107)uniform design in the experiment,seven main C-glycosylflavones fromL.gracile were taken as the investigation factors.The oxygen glucose deprivation/reoxygenation(OGD/R)induced myocardial cell damage were used as the model and cell vitality which determined by MTT were used as the main indicator.Then a further experiment for verifying the efficacy of the best proportion was conducted.The results show that the optimum proportion of C-glycosylflavones fromL.gracile is:isoorientin:swertiajaponin:afzelin:cymaroside=0.822:0.114:0.268:1.001,and it has an obviously cell protection effect(P<0.01).These results show that uniform design is reliable in seeking the optimum proportion of C-glycosylflavones from L.gracile,and this proportion has a strong protect effect on myocardial cells.
