缺铁性贫血合并心衰豚鼠模型的建立
Establishment of Guinea Pig Model of Iron Deficiency Anemia Complicated with Heart Failure
【摘要】 目的探讨缺铁性贫血合并心衰豚鼠模型的建立。方法模型组豚鼠采用低铁饲料和去离子水喂养,同时皮下注射异丙肾上腺素6周,制作缺铁性贫血合并心衰豚鼠模型。对照组豚鼠采用普通饲料和自来水喂养,同时皮下注射生理盐水6周。6周后取血测定2组豚鼠的血红蛋白含量、红细胞计数和血清铁含量;利用RM6240生物信号采集系统测定2组豚鼠的心功能和血流动力学改变情况。结果模型组豚鼠的血红蛋白含量、红细胞计数、血清铁含量、左室舒张压(LVDP)、左室收缩最大速率(+dp/dtmax)和左室舒张最大速度(-dp/dtmax)的绝对值均显著降低(P<0.01);左室舒张末压(LVEDP)显著升高(P<0.01);各项指标符合贫血和心衰的诊断标准。结论本研究成功构建缺铁性贫血合并心衰豚鼠模型,为营养学、血液病学和心血管病学的研究提供一种新的模型构建方法。
【Abstract】 Objective To investigate the establishment of a guinea pig model of iron deficiency anemia complicated with heart failure.Methods The guinea pigs in the model group were fed with low iron food and deionized water,and repeated subcutaneous injection of isoproterenol for 6 weeks,which made the guinea pig model of iron deficiency anemia complicated with heart failure.Guinea pigs in the control group were fed with normal diet and tap water,and saline was injected subcutaneously for 6 weeks at the same time.After 6 weeks,blood hemoglobin content,red blood cells,the content of serum iron of two groups were detected;the cardiac function and hemodynamic changes of two groups were detected by the RM6240 biological signal acquisition system.Results The content of hemoglobin,red blood cell count,the content of serum iron,LVDP,+dp/dtmax absolute value and-dp/dtmax absolute value of the model group were significantly decreased(P<0.01);LVEDP of the model group was significantly increased(P<0.01);the indicators met the diagnostic criteria of anemia and heart failure.Conclusion This study successfully constructed a guinea pigs model of iron deficiency anemia complicated with heart failure,and provided a new model for the study of nutriology,hematology and cardiovascular diseases.
