金雀异黄素协同雷公藤多苷干预去卵巢大鼠CIA的机制研究

【摘要】 目的:以去卵巢大鼠叠加Ⅱ型胶原诱导的类风湿关节炎(collagen-induced arthritis,CIA)为实验材料,观察金雀异黄素协同雷公藤多苷的疗效及初步药理机制,为针对绝经后女性类风湿关节炎的治疗提供理论支持及实验证据.方法:采用通用的手术法摘除大鼠卵巢,采用牛Ⅱ型胶原二次免疫的方法制备类风湿关节炎动物模型(CIA),观察金雀异黄素协同雷公藤多苷治疗雌激素低下型CIA的影响,同时与阳性药甲氨喋呤比较,评价该组合药物的药效.结果:与模型组比较,金雀异黄素协同雷公藤多苷组能较好地改善去卵巢CIA大鼠的关节肿胀及活动能力,大鼠关节炎指数及关节肿胀度均显著下降,关节组织病理程度得到明显改善.金雀异黄素协同雷公藤多苷组大鼠血清中TNF-α及IL-6水平显著降低(P<0.05),而IL-10水平呈显著升高趋势,差异有统计学意义(P<0.05);金雀异黄素协同雷公藤多苷组大鼠外周血CD4+T细胞数量/比例显著减少(P<0.01).结论:雌激素低下状态可诱导CIA的发生并加剧CIA的病理程度.补充植物雌激素可显著提高现有抗RA的药效.

【Abstract】 AIM: Rheumatoid arthritis to superposition of ovariectomized rats with collagen induced II( collagen induced arthritis,CIA) were selected as experimental materials,and the synergistic effect and preliminary pharmacological mechanism of tripterygium glycosides and genistein were observed,to provide theoretical support and experimental evidence for treatment of postmenopausal women with rheumatoid arthritis. METHODS:The rats were ovariectomized using general surgery method. The animal model of rheumatoid arthritis( CIA) was prepared using the method of secondary immune response of bovine type II collagen. The effect of genistein combined with tripterygium glycosides for the treatment of estrogen deficiency type CIA was observed,at the same time,the effect was compared with positive drug methotrexate,the curative effect of the combination of drugs was also evaluated. RESULTS: Compared with the model group,genistein combined with tripterygium glycosides group could better improve joint swelling and activity of ovariectomized CIA rats,and ratarthritis index and swelling degree were significantly decreased,the pathological degree of joint tissue was also obviously improved.The TNF- alpha and IL-6 level in the serum of rats of genistein combined with tripterygium glycosides group were significantly decreased,with statistically significant differences( P < 0. 05),and the level of IL-10 was significantly increased,with a statistically significant difference( P<0.05); The number or the ratio of peripheral blood CD4+T cell in genistein combined with tripterygium glycosides group rats was significantly reduced,with a statistically significant difference( P<0.01). CONCLUSION: Estrogen deficiency can induce the occurrence of CIA and aggravate the pathological degree of CIA. Supplement of plant estrogen can significantly improve the effectiveness of the existing anti-RA.
