家兔脑炎原虫性脑肉芽肿形成与PLCγ/PKC信号转导通路的关系
Relationship between cerebral granulomatous formation caused by Encephalitozoon cuniculi and PLCγ/PKC signal transduction pathway
【摘要】 为了研究家兔脑炎原虫性脑肉芽肿形成与PLCγ/PKC信号转导通路的关系,以及星形胶质细胞的标志性蛋白-胶质纤维酸性蛋白(GFAP)的表达,应用普通染色、特殊染色、免疫组织化学技术和Westernblot技术分别对脑炎原虫性脑组织病变进行了观察,并分析了脑炎原虫性脑肉芽肿形成与PLCγ/PKC信号转导通路关键性蛋白的关系,以及GFAP在增生性星状胶质细胞中表达。结果显示,所有患病家兔的脑组织中均有数量不等的脑肉芽肿和非化脓性脑炎变化;免疫组化染色显示,成纤维细胞生长因子(FGF)、磷酯酶Cγ(PLCγ)、磷酸化的磷酯酶Cγ(P-PLCγ)和蛋白激酶C(PKC)的表达在肉芽肿的星形胶质细胞、血管内皮细胞和成纤维细胞中明显增高,呈强阳性反应。GFAP仅在星形胶质细胞中表达,特别是增生性星形胶质细胞呈强阳性反应,被染成深棕色。用Western-blot检测,患病家兔脑组织中的FGF、PLCγ、P-PLCγ和PKC信号转导通路的关键性蛋白的表达丰度明显高于健康家兔;患病家兔脑组织中星形胶质中GFAP的表达也明显高于健康家兔。结果表明,PLCγ/PKC信号转导通路通过强大的级联反应参与了家兔脑炎原虫性脑肉芽肿的形成,GFAP在星形胶质细胞中大量表达,是脑肉芽肿中星形胶质细胞增殖的物质基础,是增生性基因被激活的表现。
【Abstract】 To study the relationship between cerebral granulomatous formation by Encephalitozoon cuniculi and PLCγ/PKC signal transduction pathway and expression of symbolic proteinglial fibrillary acidic protein in astrocytes,the common staining,special staining,immunohistochemistry and Western-blot were used to observe the morphological changes of cerebral gramuloma by E.cuniculi and analyse the relationship between the formation of cerebral gramuloma and the key proteins of PLCγ/PKC signal transduction pathway,and GFAP expression in the proliferative astrocytes.The results showed that there were a number of cerebral granuloma and nonsuppurative encephalitis in all sick rabbits.The expression of FGF,PLCγ,P-PLCγ and PKC was obviously higher in astrocytes,vascular endothelia and fibroblasts of the cerebral granuloma.These cells showed strong positive reaction and were stained as dark brown by immnohistochemistry staining.GFAP only expressed in astrocytes,especially in proliferative astrocytes.The key protein abundance of signal transduction pathway in sick rabbit’s brain tissue was much higher than that of the control by Western-blot.GFAP expression of sick rabbits’ brain tissues was apparently higher than that of the control.The conclusion is that the PLCγ/PKC signal transduction pathway is activated in process of forming cerebral granuolma by E.cuniculi via powerful cascade reaction.GFAP expression in astrocytes is a proliferative basis of astrocytes in the cerebral granuloma,and is also a manifestation of proliferative genes activated.
