壳聚糖季铵盐凝乳胶微球对O型口蹄疫VLPs免疫豚鼠的免疫增强作用

【摘要】 为了探索壳聚糖季铵盐凝乳胶微球对病毒样颗粒(virus-like particles,VLPs)的免疫增强作用,本研究利用O型口蹄疫病毒(FMDV)的病毒样颗粒作为抗原,通过Chitosan A或者加入白油后制成的混合乳Chitosan B两种壳聚糖凝胶微球与VLPs配伍免疫豚鼠,免疫后测定被免疫豚鼠的T淋巴细胞增殖情况,检测FMDV特异性抗体水平,观察计算攻毒后保护率。结果,壳聚糖季铵盐凝乳胶微球能够诱导豚鼠产生高水平的FMDV特异性抗体,T淋巴细胞增殖效果高于ISA201佐剂。此外,Chitosan B佐剂组优于Chitosan A佐剂组。上述结果说明,壳聚糖季铵盐凝乳胶微球对O型FMD VLPs具有免疫增强作用,可以诱导VLPs的体液免疫和细胞免疫应答;壳聚糖季铵盐凝乳胶微球是一种具有潜力的疫苗佐剂,可以成为FMDV疫苗的候选佐剂。

【Abstract】 To study the immune effects of the quaternized chitosan-gel microspheres on the FMDV virus-like particles(VLPs),we used the VLPs of serotype O FMDV as antigens emulsified by the chitosan microspheres(Chitosan A) or white oil chitosan microspheres(Chitosan B) to immunize guinea pigs.Tlymphocyte proliferation,FMDV specific antibodies and the protection rate of virus challenge were detected and evaluated.In result,the quaternized chitosan-gel microspheres promoted high levels of specific FMDV antibodies and T lymphocyte proliferation in guinea pigs,which was higher than that of ISA201.In addition,the result showed that Chitosan B adjuvant was better than Chitosan A adjuvant.In conclusion,the quaternized chitosan-gel microspheres can enhance immune effects of FMDV VLPs and induce both humoral and cellular immune responses.The study confirmed the quaternized chitosan-gel microspheres could be a potential vaccine adjuvant and become a new candidate of FMDV vaccine adjuvant.
