FGF2和FGFR在牦牛繁殖周期子宫的表达与定位

【摘要】 为探索成纤维细胞因子2(fibroblast growth factor,F2GF2)及成纤维细胞因子受体(fibroblast growth factor receptor,FGFR)在健康成年牦牛繁殖周期中的生物学功能,本研究选取了不同繁殖周期(发情期-卵泡期、发情期-黄体期、妊娠期-黄体期)牦牛子宫组织,采用实时荧光定量PCR(quantitative real-time PCR,RT-qPCR)和免疫组织化学方法从基因和蛋白水平分别检测不同繁殖周期牦牛子宫FGF2和FGFR的表达和分布,用积分光密度方法分析FGF2和FGFR蛋白表达水平。结果显示,不同繁殖周期牦牛子宫中FGF2mRNA表达水平为:发情期-卵泡期>发情期-黄体期>妊娠期-黄体期(P<0.01);FGFRmRNA表达水平为:发情期-黄体期>妊娠期-黄体期>发情期-卵泡期(P<0.05)。FGF2和FGFR在牦牛不同繁殖周期的子宫内膜、子宫腺及血管上皮中均有表达,子宫内膜和子宫腺为主要表达部位,阳性细胞的细胞质中表达较高。积分光密度分析结果显示,FGF2蛋白表达水平为:发情期-卵泡期子宫>发情期-黄体期子宫>妊娠期-黄体期子宫;FGFR蛋白表达水平为:妊娠期-黄体期子宫>发情期-卵泡期子宫>发情期-黄体期子宫。以上结果表明,FGF2和FGFR在不同繁殖周期的表达差异性可能与其参与早期胚胎附植及胚胎发育的调控相关,这为进一步研究FGF2和FGFR在牦牛繁殖周期中的调控作用提供了理论依据。

【Abstract】 The objective of this study was to explore the biological functions of fibroblast growth factor-2(FGF2) and fibroblast growth factor receptor(FGFR) during reproduction cycle in the uterus of yak under normal physiological conditions.The uterus of yak under different reproduction phases(oestrusluteal phase,oestrus-follicular phase and pregnancy-luteal phase)were collected,the expression and location of FGF2 and FGFR were detected by quantitative real-time PCR(q RT-PCR)and the immunohistological method,and the levels of FGF2 and FGFR protein were also measured using the integrated optical density based on the results of the immunohistological method.In result,the levels of FGF2 m RNA in the yak uterus under different reproduction phases were oestrus-follicular phase>oestrus-luteal phase>pregnancy luteal phase(P<0.01),and the levels of FGF2 mRNA were oestrus-luteal phase>pregnancy-luteal phase>oestrus-follicular phase(P<0.05).FGF2 and FGFR proteins could be detected on the endometrium,uterus gland and blood vessels of the yak uterus in different reproductions,which were mainly located in the endometrium and uterus gland;The expression of FGF2 and FGFR in positive cellswere mainly located in the cytoplasm.The levels of FGF2 showed by the integrated optical density method was oestrus-follicular phase>oestrus-luteal phase>pregnancy-luteal,while the levels of FGFR were pregnancy-luteal phase>oestrus-follicular phase>oestrus-luteal phase.The results indicated that the difference of FGF2 and FGFR during reproduction cycle might be related with its regulatory function in embryonic development and implantation, which provides information for further studies on the biological functions of FGF2 and FGFR during reproduction cycle in mammals.
