凡纳滨对虾肝胰腺组织化学过程参数的优化

【摘要】 鉴于肝胰腺组织在凡纳滨对虾生长和免疫方面的重要生理意义,以及与对虾冷藏期间品质劣变的密切相关性,本研究通过比较不同条件下石蜡切片对凡纳滨对虾肝胰腺组织HE染色结果的影响,确定其组织化学适宜参数。凡纳滨对虾肝胰腺用Carnoy固定液在室温下固定12 h,用70%,80%,95%及无水乙醇Ⅰ各脱水30min,再继续用无水乙醇Ⅱ脱水40 min,二甲苯40 min透明2次,充分浸蜡后切片(8μm),将切片经苏木精染色40 min,盐酸乙醇分化1 s,伊红复染5 min。显微观察结果显示:细胞染色均匀,细胞核呈蓝紫色,细胞质呈紫红色,背景清晰。

【Abstract】 Although the hepatopancreas tissue plays a vital physiological significance in growth and immunity of Litopenaeus vanname and it has a close relationship with the quality deterioration of shrimp during cold storage,the corresponding researches have been scarcely reported so far.The influence of paraffin-embedded section and staining parameters on the visualization of sections were thus investigated to seek the appropriate tissue preparation and processing.The results showed after fixation in Carnoy solution for 12 h and dehydration with the serial concentration of ethanol such as 70%,80%,95%,and 100% for 30 min respectively,the tissue was transpired twice with xylene for 40 min.With a sufficient immersing and embedding in paraffin wax,subsequent sectioning is performed on the Leica slicer with 8 μm in thickness and the obtained sections were immediately stained with hematine for 40 min.After differentiation in ethanol acidized with hydrochloric acid for 1 second,counterstaining of section with eosin for 5 min was conducted.Under these parameters the morphology of hepatopancreas sections observed with microscopy was clear in which the nuclear manifested bluish violet and sharp red was observed in cytoplasm parts.
