鳕鱼皮胶原蛋白肽对小鼠慢性肝损伤的保护机制

【摘要】 目的:探究鳕鱼皮胶原蛋白肽对小鼠慢性肝损伤的保护作用机制,为肝病患者功能食品的开发提供理论和试验依据。方法:采用腹腔注射CCl4建立小鼠慢性肝损伤模型,检测鳕鱼皮胶原蛋白肽对小鼠血清中转氨酶和肝组织匀浆中抗氧化酶以及丙二醛的影响;HE染色观察小鼠肝组织病理学变化;蛋白印迹法测定肝组织Bax,Bcl-2,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,JNK,P-JNK蛋白的表达情况。结果:与模型组比较,高剂量组(400 mg/kg)小鼠血清中AST、ALT活性、肝组织匀浆中MDA含量显著降低,而肝组织匀浆中SOD活性显著升高,且有显著的剂量关系;小鼠肝组织损伤情况得到明显改善;Bcl-2蛋白表达量增加,Bax,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,P-JNK蛋白表达量降低。结论:鳕鱼皮胶原蛋白肽可增强慢性肝损伤小鼠的抗氧化活性,抑制氧化应激,对慢性肝损伤有较好的保护作用。

【Abstract】 Objective: To investigate the protective effects of cod skin collagen peptide, which will provide evidences in developing function food. Methods: Chronic liver injury models were set by intraperitoneal injection CCl4 in mice. The activities of ALT, AST in serum and SOD, the contens of liver MDA were determined. Liver tissue was stained with HE for observing the effect of CSCP on mice liver histopathology. Expression of Bax, Bcl-2, Cleavage Caspase-3, Caspase-9, Cytosolic Cyt c, TNF-α, JNK, P-JNK protein were measured by western blot analysis. Reasults: Compared with model group, the groups high CSCP(400 mg/kg), ALT, AST activities in serum, MDA conten in liver were declined significantly, SOD activities significantly increased; the liver histology was improved; Expression levels of Bcl-2 improved, Bax, Cleavage Caspase-3, Caspase-9, Cytosolic Cyt c, TNF-α, JNK, P-JNK were all improved in the CSCP group. Conclusion: The cod skin collagen peptide enhanced the antioxidant activity, inhibited the oxidative stress, effectively protect the chronic liver injury by CCl4 in mice.
