连翘酯苷A对LPS诱导的鸡血液和脾脏IL-1β mRNA表达水平的影响
Effect of Forsythiaside A on IL-1β mRNA Experssion in Blood and Spleen of Chickens Induced by Lipopolysaccharide
【摘要】 试验旨在研究鸡经过脂多糖和连翘酯苷A处理后,鸡血液和脾脏IL-1β基因和蛋白水平的影响。选取240只1日龄健康AA肉鸡,并随机分为4组,经不同处理方式后,采集鸡血液和脾脏,分别采用Real-time PCR和ELISA检测IL-1β基因和蛋白水平。结果显示:与对照组相比,LPS组鸡血液和脾脏中IL-1β mRNA表达量升高(P<0.05);与LPS组比较,连翘酯苷A组鸡血液和脾脏中IL-1β mRNA表达量下降(P<0.01),并表现为一定的剂量效应。与对照组相比,LPS组脾脏中IL-1β蛋白含量显著升高(P<0.01);与LPS组比较,连翘酯苷A组鸡血清和脾脏中IL-1β蛋白含量下降(P<0.01),并表现为一定的剂量效应。表明连翘酯苷A可分别从基因和蛋白水平抑制鸡脾脏和血中IL-1β mRNA表达量与蛋白含量,进而通过影响炎症通路其他炎症因子或介导抗氧化功能调控免疫应答,减轻炎症反应。
【Abstract】 Two hundred and forty AA broilers at one day age were selected and randomly divided into four groups,treated by lipopolysaccharide(LPS) and forsythiaside A, blood and spleen were collected to detect IL-1β mRNA expression and IL-1β content level by real-time PCR and ELISA. The results showed that IL-1β mRNA expression level of spleen and blood increasd in LPS group compared with the control group(P<0.05). While forsythiaside A decreased IL-1β mRNA expression of spleen and blood compared with LPS group(P<0.05), further inhibited inflammation response and there was a dose effect. The content of IL-1β of spleen and serum were increased in LPS group compared with control group(P<0.01). While forsythiaside A could decrease inflammation response through inhibiting IL-1β content of spleen and blood in the chicken by LPS(P<0.01), and there was a dose effect. The results were suggested that forsythiaside A might decrease IL-1β mRNA expression and IL-1β content level and further through signaling pathways or antioxidant damage to regulate anti-inflammatory function.
