菊花提取物对帕金森病小鼠的保护作用及机制研究
ANTI-PARKINSONIAN ACTIVITY OF CHRYSANTHEMUM MORIFOLIUM RAMAT EXTRACTIN MICE
【摘要】 目的探讨菊花提取物(Chrysanthemum morifoliuin Ramat extract,CRE)对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)致帕金森病(PD)小鼠的保护作用及机制。方法 C57BL/6小鼠40只随机分为4组,每组10只,分别为正常组(NC)、模型组(MC)、CRE低剂量组(LC)和CRE高剂量组(HC)。LG和HC每日以CRE溶液灌胃,剂量分别为100、200mg/kg qd,NC和MC每日以等量蒸馏水灌胃,连续19 d。在第11d,除NC组外,其余组小鼠腹腔注射MPTP溶液,NC组注射等体积的生理盐水,1/d,连续5d。以爬杆法、转轴法和拉力法评价PD小鼠的行为学能力,免疫组化法(IHC)观察酪氨酸羟化酶(TH)阳性细胞数,ESI-QTOF-MS法检测纹状体多巴胺含量,采用分光光度法测定小鼠脑黑质中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)水平的变化。结果 MPTP诱导小鼠大脑发生病变,MG组行为学评分、黑质TH阳性细胞表达量、纹状体多巴胺水平及大脑黑质SOD、GSH-Px活性均显著下降,脑黑质MDA含量显著升高。与MG比较,LG、HG组行为学障碍显著改善,小鼠黑质TH阳性神经元表达、纹状体内多巴胺含量及小鼠脑黑质内SOD、GSH-Px水平均显著提高,MDA含量显著下降。结论 CRE对MPTP诱发的PD小鼠有良好的治疗作用,可能通过抑制氧化损伤起到对PD小鼠起保护作用。
【Abstract】 Objective To investigate neuroprotective effects of Chrysanthemum morifolium Ramat extract(CRE) against MPTP-induced Parkinson’s disease(PD) and its mechanism in mice. Methods C57BL/6 mice were randomly divided to 4 groups, normal control group(NC), model control group(MC) and two intervention groups which were treated with different dosages of CRE(100, 200 mg/kg, i.g.) for 19 days, respectively. PD animal model were induced by injecting MPTP and then treated with CRE accordingly. The behavioral tests, such as rotarod, pole, pulling force tests were conducted. Immunohistochemistry(IHC) test for TH positive neurons were performed to study neuroprotective effects of CRE. The levels of DA in striatum were determined by UPLC-QTOF-MS. SOD, GSH-Px and MDA levels in the striatum nigra were measured using spectrophotography. Results The score of behavioristics, mean optical density of TH and DA in the striatal tissues, the activity of GSH-Px and SOD were significantly decreased in the MC group compared with NC group,while the content of MDA was significantly increased,showing the model of PD was successfully established.The function score,mean optical density of TH and DA in the striatum tissues, activities of SOD and GSH-Px were significantly increased in the CRE treated groups compared with the MC group,while the content of MDA was significantly decreased.Conclusion This study indicates Chrysanthemum morifoliuin Ramat extract has significant therapeutic effect on MPTP induced PD,which suggests its potential as a new candidate in treating PD.
