维生素D3联合抗坏血酸对豚鼠结肠炎的改善作用

【摘要】 目的探讨维生素D3(VD3)联合抗坏血酸(VC)对豚鼠溃疡性结肠炎的作用。方法将40只雄性豚鼠按体质量随机分为5组,每组8只:正常组[N,120mg VC+2.8μg VD3/(kg·d)]、模型组[M,120mg VC+2.8μg VD3/(kg·d)]、高VC低VD组[HCLD,240mg VC+1.4μg VD3/(kg·d)]、中VC低VD组[MCLD,120mg VC+1.4μg VD3/(kg·d)]和低VC低VD组[LCLD,8mg VC+1.4μg VD3/(kg·d)]。每组均进食缺乏VD和VC的特殊饲料,采用改良的豚鼠灌胃方法给予各组相应剂量的VD3和VC。灌胃7w后,N组继续饮用蒸馏水,其他4组饮用3%葡聚糖硫酸钠溶液3d,造模期间,灌胃情况同前。结果 N组体质量、粪便和精神状态正常,评分明显低于其他4组,血清中25-(OH)D3、1,25-(OH)2D3显著升高,C反应蛋白(CRP)、白介素-6(IL-6)和核转录因子κB(NF-κB)明显降低;与LCLD组相比,HCLD组大体形态改善,组织学损伤较轻,疾病活动指数评分较低,血清中25-(OH)D3、1,25-(OH)2D3显著升高,CRP、IL-6和NF-κB明显降低;M组评分显著低于LCLD组,其25-(OH)D3、1,25-(OH)2D3显著高于LCLD组,CRP、IL-6和NF-κB较HCLD、MCLD和LCLD组明显降低,且均有统计学意义(P<0.05)。结论维生素D3缺乏时,较高剂量的VC对豚鼠溃疡性结肠炎的防治效果更明显。

【Abstract】 Objective To investigate the effect of vitamin D3 combined with ascorbic acid on ulcerative colitis in guinea pigs. Methods Forty male Dunkin-Hartley guinea pigs were randomly divided into 5 groups according to their weight, with 8 guinea pigs in each group:normal group(N, 120 mg VC+2.8μg VD3/(kg·d)), model group(M, 120 mg VC+2.8μg VD3/(kg·d)), high VC and low VD group(HCLD, 240 mg VC+1.4μg VD3/(kg·d)), middle VC low VD group(MCLD, 120 mg VC+1.4μg VD3/(kg·d)) and low VC low VD group(LCLD, 8mg VC+1.4μg VD3/(kg·d)). Guinea pigs in each group were fed a special diet without vitamin D and vitamin C, and the corresponding doses of VC and VD were given by gavage. After 7 weeks, the normal group continued drinking distilled water, and the other 4 groups were treated with 3% dextran sodium sulfate solution for 3 days. Within three days, the intervention was unchanged. Results N group had normal body weight, stool and mental status, and scores of N group were significantly lower than those in the other 4 groups. The serum 25-(OH)D3, 1,25-(OH)2D3 increased significantly. C-reactive protein(CRP), interleukin-6(IL-6) and nuclear factor kappa B(NF-κB) were significantly decreased. Compared with LCLD group, gross morphology was improved in the HCLD group. Meanwhile, histological damage was less severe, disease activity index score was lower, serum 25-(OH)D3, 1,25-(OH)2D3significantly increased, and CRP, IL-6 and NF-kappa B significantly decreased. The score of M group was significantly lower than that of the LCLD group, while 25-(OH)D3, 1,25-(OH)2D3 was significantly higher than that of LCLD group, and CRP, IL-6 and NF-kappa B was significantly lower than that of HCLD, MCLD and LCLD groups(P<0.05). Conclusion The higher dose of VC was more effective in the prevention and treatment of ulcerative colitis in guinea pigs when vitamin D3 was deficient.
