滇重楼中重楼皂苷和重金属含量的相关性研究

【摘要】 目的:测定滇重楼中4种重楼皂苷和8种重金属元素的含量,探讨上述2种质量评价指标间的相关性。方法:采用电感耦合等离子体发射光谱法测定滇重楼根茎内铅、镉、铜、铬、钡和锑的含量;原子荧光光谱分析法测定汞、砷的含量;高效液相色谱法测定重楼总皂苷(包括重楼皂苷Ⅰ、重楼皂苷Ⅱ、重楼皂苷Ⅵ和重楼皂苷Ⅶ)的含量,并采用SPSS 23.0软件对数据进行相关性分析。结果:滇重楼根茎中铅、镉、铜、汞、砷、铬、钡和锑的平均含量分别为20.433、50.020、67.852和280.491μg/kg,1.632、1.982、9.356和15.875 mg/kg;重楼总皂苷含量为0.811%,其中重楼皂苷Ⅰ占总皂苷量的72.4%。相关性分析结果显示,重楼总皂苷的含量与8种重金属的含量无显著相关性,但重楼皂苷Ⅰ的含量与镉的含量呈极显著负相关,与铜、锑的含量呈极显著正相关,与砷的含量呈显著正相关。结论:滇重楼中活性成分的含量与重金属元素的含量有一定相关性,可为建立滇重楼全面质量评价体系提供数据依据。

【Abstract】 OBJECTIVE:To determine the content of 4 kinds of polyphyllins and 8 kinds of heavy metals in Paris polyphylla var.yunnanensis,so as to probe into the correlation between the indicators of quality evaluation.METHODS:The content of Pb,Cd,Cu,Cr,Ba and Sb were determined by ICP-AES;the content of Hg and As were determined by AFC;the content of Rhizoma Paridis total saponin(parissaponinⅠ,parissaponin Ⅱ,parissaponin Ⅵ and parissaponin Ⅶ)were determined by HPLC;the data were analyzed by SPSS 23.0 software.RESULTS:The average contents of Pb,Cd,Cu,Hg,As,Cr,Ba and Sb in rhizome of Paris polyphylla var.yunnanensis were 20.433 μg/kg,50.020 μg/kg,67.852 μg/kg,280.491 μg/kg,1.632 mg/kg,1.982 mg/kg,9.356 mg/kg and 15.875 mg/kg respectively.The total amount of polyphyllins was 0.811%,of which 72.4% was parissaponin Ⅰ.The correlation analysis indicated that the content of total amount of polyphyllins was not significantly correlated with the content of 8 kinds of heavy metals,yet there was significant negative correlation between the content of parissaponin Ⅰ and Cd,and significant positive correlation with the content of Cu and Sb,as well as significant positive correlation with the content of As.CONCLUSIONS:There is certain correlation between the content of the active components of Paris polyphylla var.yunnanensis and heavy metal elements,which can provide data support for the development of the comprehensive quality evaluation system.
