甘草苷对UVB诱导HaCaT细胞光老化的保护作用及机制研究

【摘要】 目的研究甘草苷对中波紫外线(UVB)诱导人皮肤角质形成细胞(HaCaT)光老化的保护作用及机制。方法用不同浓度的甘草苷作用于HaCaT细胞,噻唑蓝比色(MTT)法筛选甘草苷的安全有效浓度;用30 m J/cm2的UVB照射HaCaT细胞,建立光老化模型。用10-7、10-6、10-5 mol/L的甘草苷处理甘草苷+UVB组细胞24 h,MTT法检测甘草苷对光老化HaCaT细胞增殖率的影响;试剂盒检测HaCaT细胞中谷胱甘肽(GSH)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量;蛋白免疫印记法检测HaCaT细胞中白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)蛋白表达量。结果 10-7、10-6、10-5 mol/L甘草苷对HaCaT细胞无明显的促增殖与抑制作用(P>0.05),为最佳安全有效浓度。与空白组相比,UVB组细胞增殖率显著降低,GSH、CAT活性显著降低,MDA含量显著升高,IL-1β、IL-6、TNF-α蛋白表达量显著升高(P<0.01)。与UVB组相比,10-7、10-6、10-5mol/L甘草苷+UVB组细胞增殖率显著升高,GSH、CAT活性明显上升,MDA含量显著降低,IL-1β、IL-6、TNF-α蛋白表达量显著降低(P<0.05或P<0.01)。结论甘草苷对UVB诱导的HaCaT细胞光老化有保护作用,其机制与其提高抗氧化酶活性,抑制炎性因子的产生有关。

【Abstract】 Objective To investigate the protective effects and the mechanism of liquiritin on photoaging human keratinocytes(HaCaT)cells induced by ultraviolet B(UVB).Methods The different concentrations of liquiritin were used on HaCaT cells,the best effective and safe concentration of liquiritin was determined by methyl thiazolyl tetrazolium(MTT).The photoaging model was set up by the irradiation of UVB at irradiation dose of 30 m J/cm2.It was used to treat liquiritin+UVB group cells 24 h with 10-7,10-6,10-5mol/L,the effects of liquiritin on the proliferation rate of photoaging model HaCaT cells were detect by MTT.The activity of glutathione(GSH),catalase(CAT)and the content of malondialdehyde(MDA)were detected by the corresponding kits.The detection of interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)protein expression were detected by Western blot.Results The liquiritin with the concentrations of 10-7,10-6,10-5mol/L had no obvious effect on the proliferation and inhibition of HaCaT cells(P>0.05),which were the best safe and effective concentrations.Compared with the blank group,the cell proliferation rate of the UVB group was significantly decreased,the activity of GSH and CAT was significantly decreased,and the content of MDA was significantly increased,the expression of IL-1β,IL-6 and TNF-αprotein were increased(P<0.01).Compared with the UVB group,the cell proliferation rates of the(10-7,10-6,10-5)mol/L liquiritin+UVB group were significantly increased,the activity of GSH and CAT was significantly increased,the content of MDA was significantly de creased,the expression of IL-1β,IL-6 and TNF-αprotein were significantly decreased(P<0.05 or P<0.01).Con-clusion Liquiritin can protect HaCaT cells photoaging,which mechanism is related to the improvement of antioxidant enzyme activity and inhibition of in-flammatory cytokines.
