【摘要】 目的 研究过表达钙粘蛋白P11对人乳腺癌细胞增殖及迁移能力的影响。方法 通过转染实验在MCF-7细胞株中构建钙粘蛋白P11过表达细胞株MCF7-pKvenus-P11和阴性对照细胞株MCF7-pKvenus,行Western blot检测过表达细胞株中钙粘蛋白P11蛋白表达,验证钙粘蛋白P11过表达模型是否构建成功,行CCK-8细胞活性实验检测MCF7-pKvenus-P11的增殖活性是否变化,并行细胞划痕实验及transwell侵袭实验检测MCF7-pKvenus-P11的迁移能力是否发生改变。结果 Western blot显示,过表达细胞株MCF7-pKvenus-P11中钙粘蛋白P11的蛋白表达明显升高(P<0.05),提示钙粘蛋白P11过表达细胞株MCF7-pKvenus-P11建立成功;钙粘蛋白P11过表达后MCF-7增殖及迁移能力明显增强(P<0.05)。结论钙粘蛋白P11可促进人乳腺癌细胞的增殖及迁移。更多还原

【Abstract】 Objective To explore the effects of overexpression cadherin P11 on migration and proliferation ability of human breast cancer cells.Methods P11-overexpression cell lines MCF7-pKvenus — P11 and negative control cell lines MCF7-pKvenus were established in MCF-7 cell lines through transfection experiment.The expression of P11 was tested by western blot to measure transfer efficiency.Proliferation activity of MCF7-pKvenus-P11 was detected by CCK-8 cell activity experiment.Migration ability of MCF7-pKvenus-P11 was detected by wound healing assay and transwell invasion experiment.Results Western blot showed that MCF7-pKvenus-P11 contained more P11 than MCF7-pKvenus(P <0.05),which meant successful establishment of P11-overexpression cell lines MCF7-pKvenus-P11.P11-overexpression could promote the proliferation and migration ability of breast cell line(P < 0.05).Conclusion P11 can promote the proliferation and migration ability of human breast cancer cells.更多还原
