Ⅱ型胶原酶消化分离法和组织块直接培养法体外培养人软骨细胞的效果
Comparison of Human Chondrocytes Cultured in Vitro by Type Ⅱ Collagenase Digestion Method and Explant Culture Method
【摘要】 目的比较Ⅱ型胶原酶消化分离法和组织块直接培养法体外培养人软骨细胞的效果。方法取人外伤性截肢及原发性骨关节炎患者的膝关节软骨,无菌条件下将软骨剪成1 mm×1 mm×1 mm左右的碎块,分别采用Ⅱ型胶原酶消化法和组织块直接培养法分离培养人关节软骨细胞,进行原代及传代培养,观察软骨细胞的生长情况。结果采用Ⅱ型胶原酶消化法分离培养可获得较多软骨细胞,细胞形态典型,增殖较快。组织块直接培养法获得的软骨细胞数量较少,增殖慢。结论两种不同方法培养后,均可获得人软骨细胞的生长,Ⅱ型胶原酶消化分离培养法明显优于组织块直接培养法。
【Abstract】 Objective To compare the effect of the type Ⅱ collagenase digestion method and tissue block direct culture method on human chondrocytes cultured in vitro. Methods We took sterile knee cartilage tissue samples from people with traumatic amputation or primary osteoarthritis, cut them into 1 mm × 1 mm × 1 mm pieces under sterile condition, then applied primary and subculture of human articular chondrocytes through the type Ⅱ collagenase digestion method or tissue block direct culture method. We observed the growth of chondrocytes of both groups. Results Chondrocytes were more easier to be obtained, more morphologically typical and proliferated faster in group using type Ⅱ collagenase for isolation and culture. The type Ⅱ collagenase method was significantly superior to the tissue direct culture method. Conclusion After cell culture, growth of human chondrocytes were eligible for both groups. The type Ⅱ collagenase digestion method is significantly better than tissue block direct culture method.
