基于系统药理学探索痛泻要方治疗肠易激综合征的分子机制

【摘要】 目的:建立痛泻要方治疗肠易激综合征(IBS)的"化合物-靶点-通路"的网络,研究其多成分、多靶点、多途径的作用机制。方法:利用TCMSP数据库为基础查找痛泻要方的化学成分,利用Drug Bank数据库检索作用靶点,构建药物-成分-靶点数据库;在Pub Med Gene数据库中检索疾病相关基因,通过Kyoto Encyclopedia of Genes and Genomes(KEGG)数据库对获取的靶点进行相关通路分析,利用Cytoscape软件构建痛泄要方的"化合物-靶点-通路-疾病"网络图。结果:获得痛泻要方中有效成分42种,相关作用靶点18个,涉及35条信号通路。结论:痛泻要方可能通过作用于免疫、炎症、内分泌系统的相关蛋白靶点及通路,起到治疗IBS的作用。

【Abstract】 Objective: To establish the network of "compound-target-pathway"in the treatment of irritable bowel syndrome( IBS) in Tongxieyaofang,and study its multi-component,multi-target and multi-path mechanism. Methods: The drug-constituent-target database was constructed by using the Drug Bank database,and the disease-related genes were searched in the Pub Med Gene database. The genes of the disease-related genes were searched by the Kyoto Encyclopedia of Genes and Genomes( KEGG database was used to analyze the acquired pathways,and Cytoscape software was used to construct the"compound-target-pathway-disease"network. Results: There were 42 effective components in pain-relieving recipe and 18 relatedrelated targets,involving 35 signal pathways. Conclusion: Tongxieyaofang may play a role in the treatment of IBS through the role of immune,inflammation,endocrine system related protein targets and pathways.
