【摘要】 [目的]观察糖尿病胃动力障碍大鼠胃窦平滑肌组织中磷酯酶Cβ亚型(PLCβ)和蛋白激酶C(PKC)的表达情况.[方法]取Wistar大鼠,腹腔注射给予STZ制作糖尿病大鼠模型,利用RM-6240四道生理信号记录仪观察大鼠胃窦环行平滑肌的自发性收缩运动情况,将自发性收缩运动减弱的大鼠确定为糖尿病胃动力障碍模型.通过免疫组织化学方法检测PLCβ和PKC蛋白在糖尿病胃动力障碍大鼠胃窦平滑肌组织中的表达水平.[结果]与正常对照组比较,糖尿病胃动力障碍大鼠胃窦平滑肌组织中PLCβ和PKC蛋白表达水平均明显降低(P<0.05).[结论]糖尿病大鼠发生胃动力障碍原因认为可能与胃窦平滑肌中PLCβ和PKC蛋白表达水平下调有关系.更多还原

【Abstract】 OBJECTIVE To observe the expressions of phospholipase C(PLC)βsubtype and protein kinase C(PKC)in gastric antrum smooth muscle of rats with diabetic gastric motility disorders.METHODS Wistar rats were taken to establish the diabetes model by intraperitoneal injection of STZ.RM-6240 four channels physiological recorder was used to observe the spontaneous contractions of rat gastric antrum circular smooth muscle.The model of diabetic gastric motility disorders was determined by the rats with hypomotility of spontaneous contractions.Immunohistochemistry was used to detect the expressions of PLCβsubtype and PKC in gastric antrum smooth muscle of rats with diabetic gastric motility disorders.RESULTSCompared with the rats in the control group,the expressions of PLCβsubtype and PKC in gastric antrum smooth muscle of rats with diabetic gastric motility disorders were significantly decreased(P<0.05).CONCLUSION Gastric motility disorders of diabetes rats might be correlated with the down-regulation of PLCβsubtype and PKC in gastric antrum smooth muscle.更多还原
