外源性硫化氢供体硫氢化钠改善慢性应激致抑郁大鼠神经元损伤的研究

【摘要】 目的探讨外源性硫化氢供体硫氢化钠对慢性应激致大鼠抑郁模型神经元损伤的影响,为抑郁症的防治提供新的干预靶点。方法将48只SD大鼠按随机数字表法分为正常组、空白对照组、抑郁组、硫氢化钠组,每组12只。除去建模中死亡的大鼠,最后每组入选6只大鼠。慢性应激建立大鼠抑郁模型,硫氢化钠组大鼠给予硫氢化钠10 mg/kg,抑郁组、空白对照组给予等量生理盐水腹腔注射,每周1次。收集大鼠海马组织及血浆,采用比色法检测海马及血浆中硫化氢水平,逆转录-聚合酶链反应和免疫印迹法检测海马组织中胱硫醚β合成酶(CBS)m RNA及蛋白表达,苏木精-伊红和尼氏染色观察海马结构和神经元形态。结果抑郁组大鼠海马及血浆硫化氢水平及海马组织中CBS m RNA及蛋白表达量均明显低于空白对照组、硫氢化钠组,差异均有统计学意义(P<0.05)。抑郁组大鼠海马区尼氏体明显减少,锥体细胞排列紊乱,细胞核消失;硫氢化钠组大鼠海马尼氏体的缺失及锥体细胞排列紊乱较抑郁组明显改善。结论内源性硫化氢可能参与了慢性应激致大鼠抑郁的发生,而外源性硫化氢供体硫氢化钠能显著改善抑郁大鼠海马区神经元损伤。

【Abstract】 Objective To explore the effects of exogenous hydrogen sulfide(H2 S) donor sodium hydrosulfide(Na HS) on neurons damage of depression rats caused by chronic stress,and provide new targets for preventing and treating depression.Methods Forty-eight Sprague Dawley(SD) rats were divided into the normal group,the blank control group,the depression group and the Na HS group according to random number table method in March 2016,12 cases in each group,but finally 6 case in each group after removing death SP rats in the process of modeling. SD rats depression model was established by chronic stress,the Na HS group was administered with sodium hydrosulfide 10 mg/kg by introperitoneal injection,once a week;the depression group and the blank control group were administered with equivalent normal saline by intraperitoneal injectio,once a week. The hippocampus tissues and plasma of SD rats were collected to detect the expression of cystathionine-synthase(CBS) m RNA and protein in hippocampus tissues by RT-PCR and Western blotting,to observe the hippocampus formation and morphology of neurons by HE staining and Nissl′ s staining,and to determine the level of H2 S in hippocampus tissues and plasma by colorimetry.Results The levels of H2 S in plasma and hippocampus in the depression group were obviously lower than those in the blank con-trol group,and the expression of CBS m RNA and protein in the hippocampus tissue were evidently lower than those in the blank group and the Na HS group,with statistically significant difference(P<0.05). In the depression group,Nissl body in hippocampus of SD rats reduced distinctly,pyramidal cells showed irregular arrangement and the cellular nucleus vanished;while in the Na HS group,the deficiency of Nissl body in hippocampus and the irregular management of pyramidal cells were improved more promi-nently than those in the depression group. Conclusion Endogenous H2 S may be involved in the depression induced by chronic stress in SD rats,while H2 S donor Na HS can attenuate the damage of hippocampal neurons in depressed rats.
