叶酸对硫酸铝钾诱发大鼠脑组织氧化应激损伤及胆碱能系统的影响
Effect of folic acid on the brain oxidative stress injury and cholinergic system
【摘要】 目的探讨叶酸对硫酸铝钾诱发大鼠脑组织氧化应激损伤及胆碱能系统的影响。方法选择健康3月龄SD大鼠50只,将其分为对照组、模型组、3.2 mg/kg叶酸治疗组、1.6 mg/kg叶酸治疗组、0.8 mg/kg叶酸治疗组,每组10只。采用灌胃方式给予大鼠十二水硫酸铝钾100 mg/kg造模,造模同时开始分别给予大鼠不同剂量叶酸连续灌胃4周后处死所有大鼠,取脑组织,测定脑组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量及谷胱甘肽过氧化物酶(GSH-Px)、乙酰胆碱酯酶(ACh E)、胆碱乙酰转移酶(Ch AT)活性变化。结果与对照组比较,模型组大鼠脑组织SOD活性明显降低,MDA含量显著升高,GSH-Px活性明显降低,脑组织匀浆的ACh E活性升高,Ch AT活性降低,差异均有统计学意义(P<0.05)。与模型组比较,叶酸治疗组(3.2、1.6 mg/kg)大鼠脑组织SOD活性升高,MDA含量明显降低,ACh E活性降低,Ch AT活性升高,差异均有统计学意义(P<0.05)。结论叶酸具有清除氧自由基和抗氧化功能,可能通过提高SOD活性,降低MDA含量来改善痴呆,提高脑组织中胆碱能神经递质的活性,从而保护神经细胞。
【Abstract】 Objective To explore the effect of folic acid on the rats′ brain oxidative stress and cholinergic system induced by aluminum potassium sulfate. Methods A total of 50 three-months old healthy SD rats were divided into the control group,model group,3.2,1.6,0.8 mg/kg folic acid treatment group,10 rats in each group. The model group rats by gavage with100 mg/kg aluminum potassium sulfate,at the same time,starting gavage separately and continuously for 4 weeks with folic acid,all rats were sacrificed to get the brain tissue,determined the activity of superoxide dismutase(SOD),content of malondialdehyde(MDA),and the activity of glutathione peroxidase(GSH-Px),acetylcholinesterase(ACh E) and choline acetyltransferase(Ch AT). Results Compared with the control group,the activity of SOD in the brain tissue of model group was significant decreased,the content of MDA was significant increased,the activity of GSH-Px significant was decreased,the activity of ACh E was increased,the activity of Ch AT was decreased,the differences were statistically significant(P<0.05). Compared with model group,the activity of SOD in the brain tissue of folic acid(3.2,1.6 mg/kg) treatment group were significant increased,and the content of MDA were significant decreased;the activity of ACh E were decreased,the activity of Ch AT were increased,the differences were all statistically significant(P<0.05). Conclusion The folic acid has a function of oxygen free radicals scavenging,as well as has the effects of antioxidation. It can ameliorate dementia by enhancing the activity of SOD activity and reducing the content of MDA,and the activities of cholinergic neurotransmitters in the brain tissue is improved to protect nerve cells.
