沙立度胺抑制Wistar大鼠动脉粥样硬化斑块的形成

【摘要】 目的观察沙立度胺对Wistar大鼠动脉粥样硬化斑块的影响。方法将40只雄性Wistar大鼠采用完全随机设计法分为正常对照组(A组)、动脉粥样硬化组(B组)、小剂量沙立度胺组(C组)和大剂量沙立度胺组(D组),每组10只。A组给予普通饲料喂养12周;B、C、D组均给予高脂饲料喂养12周。在喂养开始时给予腹腔注射维生素D360万U/kg(A组注射等量生理盐水),以后每隔1个月注射1次,并于实验开始后第7天行大鼠主动脉内膜球囊损伤术,建立动脉粥样硬化动物模型。C、D组给予沙立度胺灌胃给药,剂量分别为10、30 mg/(kg·d),第12周末处死40只大鼠,取血分离血清,检测三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)等血脂指标,取整条主动脉,行苏木精-伊红染色观察斑块病变程度,检测主动脉内、中膜厚度。结果 (1)B、C、D组大鼠血清TG、TC、LDL-C等血脂指标均明显高于A组,HDL-C水平均明显低于A组,差异均有统计学意义(P<0.05);B、C、D组大鼠血清TG、TC、LDL-C、HDL-C等血脂指标比较,差异均无统计学意义(P>0.05)。(2)C组大鼠斑块内脂质含量、泡沫细胞及炎症细胞数量、内膜厚度均明显低于B组,中膜厚度明显高于B组,差异均有统计学意义(P<0.05);D组大鼠病变程度明显轻于C组,可见少量泡沫细胞及炎症细胞浸润,内膜厚度明显低于C组,中膜厚度明显高于C组,差异均有统计学意义(P<0.05)。结论沙立度胺可抑制Wistar大鼠动脉粥样硬化斑块的形成,与脂质代谢调节无关。且其抑制动脉粥样斑块形成的作用可能与剂量呈正相关。

【Abstract】 Objective To observe the effect of thalidomide on the atherosclerotic plaque in Wistar rat. Methods Forty Wistar male rats were randomly divided into the normal control group(A),atherosclerosis group(B),low dose thalidomide group(C) and high dose thalidomide group(D) according to the complete random design method. The group A was fed with normal fodder for 12 weeks and the other three groups were fed with high fat fodder. At the beginning of feeding,vitamin D3 600 000 IU/kg was intraperitoneally injected(same volume of normal saline in the group A),afterward once injection every other month,and the rat aortic intima balloon injury operation was performed on 7 d after the start of experiment for establishing the atherosclerotic animal model. The group C and D were given thalidomide gavage,the doses were 10,30 mg/(kg·d) respectively. All 40 rats were executed at the end of 12 weeks. And then the blood sample was collected and separated for detecting the lipid indexes such as TG,TC,LDL-C and HDL-C. Furthermore,the whole aorta was taken for conducting the HE staining,observing the degree of plaque lesion and detecting the thickness of aortic intima and tunica media. Results(1) The blood-lipid indexes of TG,TC and LDL-C in the group B,C and D were significantly higher than those in the group A,the difference was statistically significant(P<0.05),but the blood lipid indexes of TG,TC,LDL-C and HDL-C had no statistical difference among the group B,C and D(P>0.05).(2)The lipid content,number of foam cells and inflammatory cells of plaque and thickness of aortic intima in the group C were significantly lower than those in the group B,the tunica media thickness was significantly higher than that in the group B,the differences were statistically significant(P<0.05). The lesion degree in the group D was slighter than that in the group C,showing a small amount of foam cells and inflammatory cells infiltration,the intima thickness was significantly lower than that in the group C and the tunica media thickness was significantly higher than that in the group C,the differences were statistically significant(P<0.05).Conclusion Thalidomide can inhibit the formation of atherosclerotic plaque in Wistar rat,which has no relation with lipid metabolism regulation. And its effect for inhibiting arterial atherosclerotic plaque formation may be positively correlated with the dose.
