红景天苷对胶质瘤U87-MG细胞增殖和侵袭能力的影响

【摘要】 目的观察红景天苷对胶质瘤U87-MG细胞增殖和侵袭能力的影响,探讨其诱导U87-MG细胞凋亡机制。方法不同浓度红景天苷和喜树碱加入U87-MG细胞中,作用24 h后,MTT法测定U87-MG细胞增殖,流式细胞术检测U87-MG细胞凋亡率,Transwell法检测U87-MG细胞侵袭能力,间接荧光标记法测定细胞内活性氧(ROS)水平,Western blot检测U87-MG细胞Caspase-3、Bcl-2、Bax、E-cadherin、N-cadherin和基质金属蛋白酶-9(MMP-9)的表达。结果与空白对照组比较,各给药组U87-MG细胞生长抑制作用明显,10、50、100μg/m L红景天苷作用U87-MG细胞后,细胞侵袭能力显著下降,10、50、100μg/m L红景天苷作用U87-MG细胞48 h后均能诱导U87-MG细胞凋亡;ROS水平与红景天苷浓度呈正相关;10、50、100μg/m L红景天苷上调Caspase-3、Bax和E-cadherin的表达,下调Bcl-2和N-cadherin和MMP-9的表达。结论红景天苷可诱导U87-MG细胞凋亡、抑制U87-MG细胞的侵袭能力。

【Abstract】 Objective To investigate the effects of salidroside on proliferation and invasive ability of glioma U87-MG cells; To discuss the its mechanism to induce apoptosis of U87-MG cells. Methods U87-MG cells were cultured in vitro for 24 h under different concentrations of salidroside and camptothecin. The proliferation of U87-MG cells was detected by MTT assay. The apoptosis rate of U87-MG cells was detected by flow cytometry. Transwell assay was used to detect the invasive ability of U87-MG cells. ROS was detected by indirect fluorescent labeling. The expressions of Caspase-3, Bcl-2, Bax, E-cadherin, N-cadherin and matrix metalloproteinase-9(MMP-9) in U87-MG cells were detected by Western blot. Results Compared with the blank control group, U87-MG cells had significant inhibitory effect on the growth of U87-MG cells in each administration group, and the invasive ability of U87-MG cells was significantly reduced after 10, 50, 100 μg/m L salidroside was intervened, and 10, 50, 100 μg/m L salidroside for 48 h for U87-MG cells could induce apoptosis of the cells; the level of ROS was positively correlated with the concentration of salidroside; 10, 50, 100 μg/m L salidroside up-regulated the expressions of Caspase-3, Bax and E-cadherin, and down-regulated the expressions of Bcl-2, N-cadherin and MMP-9. Conclusion Salidroside can induce apoptosis of U87-MG cells and inhibit the invasive ability of U87-MG cells.
