藏鸡源食窦魏斯氏菌的抑菌特性研究

【摘要】 为了解藏鸡源益生菌的抑菌特性,选用10只体质健康的1月龄藏鸡作为益生菌的分离来源。以肠炎沙门菌(Salmonella enterica)为指示病原菌,通过琼脂扩散试验对分离菌株进行抑菌特性判定,共筛选到6株具有抑制肠炎沙门菌生长繁殖的菌株,抑菌圈直径均在8~28 mm之间,其中从小肠黏膜中分离到的食窦魏斯氏菌(Weissella cibaria)的抑菌圈直径高达276 mm,明显高于其他菌株和常用抗生素。将该菌株命名为WT1,并对WT1的抑菌特性和生物学活性进行深入研究。结果显示:WT1所产细菌素作用肠炎沙门菌8 h后,肠炎沙门菌大量死亡;基因序列分析显示,WT1益生基因entA;WT1具有良好的热稳定性和耐酸性。研究表明,藏鸡源食窦魏斯氏菌具有良好的抑菌特性和生物学活性,可以用于家禽肠炎沙门菌感染的治疗,在一定程度上缓解抗生素的负面影响。

【Abstract】 The present study was conducted to explore more meaningful treatment strategies and characterize bacteriostatic action of probiot-ics in intestinal flora of Tibetan chickens. A total of 6 strains capable of inhibitingSalmonella entericawere selected, and the diameters of in-hibition zone ranged from 8 mm to 28 mm. Especially, the diameters of inhibition zone ofWeissella cibariastrain WT1 was found to be thehighest(27 6 mm). Our results showed that WT1 produced bacteriocins with inhibitory activity against indigenousS. enterica, and the valueof OD260 reached 3 86 after 8 h. In addition, the bacteriostasis of WT1 retained stable with the treatment of heat and kept active after beingexposed to acidulous and neutral conditions. Furthermore, the WT1 was positive for the primers ofentA in the PCR. Our finding indicatedthatWeissella cibariacould provide a rich source for the discovery of novel antimicrobial substance with potential benefit in inhibiting Salmo-nella infection.
