鸡传染性法氏囊病病毒超强毒江苏分离株的鉴定及其致病特征分析

【摘要】 对2009年采自江苏南京的疑似鸡传染性法氏囊病(IBD)的病料进行了鸡胚分离纯化,通过琼脂扩散、红细胞凝集、血清中和试验、RT-PCR等试验确定分离毒NJ09是传染性法氏囊病病毒(IBDV)。对其VP2基因序列分析显示:NJ09与美国CAHFSK669分离株及中国的B-SDRZ、SD10LY01分离株亲缘关系较近,共同聚类于一个超强毒株分枝,说明NJ09是一个IBDV超强毒株(vvIBDV)。致病性测定结果显示:NJ09对鸡胚的半数致死量(ELD50)为1065/02 mL,对60日龄SPF鸡的攻毒发病率和致死率分别达100%和60%。免疫原性测定结果显示:NJ09的灭活疫苗有较好的保护效果,血清中和抗体值达156 log2。上述结果表明,NJ09是一株vvIBDV,致病力强,免疫原性好,在后续IBDV疫苗研发中是潜在的高效种毒来源。

【Abstract】 Chicken bursal samples with typical infectious bursal disease(IBD) symptoms were collected from Nanjing, Jiangsu Province.One virus strain of infectious bursal disease virus(IBDV) named NJ09 was isolated and confirmed by agarose diffusion, hemagglutination,serum neutralization test and RT-PCR amplification. Phylogenetic analysis based on the nucleic acid identities of VP2 revealed that NJ09 shared the highest identities with the US isolate CAHFSK669 and two China isolates(B-SD-RZ, SD10LY01) and clustered in a very viru-lent infectious bursal disease virus(vvIBDV) branch, which indicated NJ09 was an isolate of vvIBDV. The result of inoculation test on SPFembryo showed NJ09 could cause the chicken embryo death and the titer of the virus could reach as high as 106.5ELD50/0 2mL. Also, theinfection rate and the lethality rate of NJ09 were as high as 100% and 60% in 60-day-old chickens, respectively. After inactivated, NJ09 al-so showed good immunogenicity and the titer for serum neutralizing antibody was 15 6 log2 All these results showed NJ09 was an isolate ofvvIBDV, with high pathogenicity and good immunogenicity, which indicated NJ09, as an efficient seed virus, had the potential to be appliedon the research and development of vaccine against IBDV in the future.
